摘 要:目的探讨贝伐珠单抗对高表达HER-2/neu卵巢癌小鼠的治疗作用。方法选择雌性健康小鼠76只,将人卵巢癌SKOV3细胞接种于小鼠建立模型,然后随机分成两组各38只。对照组尾静脉注射顺铂,观察组尾静脉注射顺铂同时腹腔注射贝伐珠单抗,比较两组SKOV3细胞生长抑制率,观察两组小鼠瘤体的生长抑制情况,对比两组小鼠的HER-2/neu标记指数、p53标记指数。结果观察组细胞生长抑制率高于对照组,差异有统计学意义(P<0.01)。观察组的肿瘤抑制率(95.54±6.57)%高于对照组肿瘤抑制率(70.62±5.11)%,差异有统计学意义(P<0.01)。观察组小鼠的HER-2标记指数、p53标记指数都低于对照组,差异均有统计学意义(P<0.01)。结论贝伐珠单抗能抑制高表达HER-2/neu卵巢癌肿瘤细胞的生长,作用机制可能与贝伐珠单抗能够调控p53基因及HER-2/neu基因表达有关。Objective To explore the therapeutic effect of bevacizumab on ovarian cancer mice with high expression of HER- 2 / neu.Methods A total of 76 healthy female mice were selected,then human ovarian cancer SKOV3 cells were inoculated into the mice to establish mouse model,then the mice were divided into two groups,38 mice in each group. The mice in control group were treated by tail vein injection of cisplatin,while the mice in observation group were treated by tail vein injection of cisplatin combined with intraperitoneal injection of bevacizumab. The growth inhibition rates of SKOV3 cells in the two groups were compared; the inhibition situations of tumor growth in the two groups were observed,HER-2 / neu labeling indexes and p53 labeling indexes in the two groups were compared. Results Cell growth inhibition rate in observation group was statistically significantly higher than that in control group( P0. 01). The inhibition rate of tumor in observation group was( 95. 54 ± 6. 57) %,which was statistically significantly higher than that in control group( 70. 62 ± 5. 11) %( P〈 0. 01). HER-2 / neu labeling index and p53 labeling index in observation group were statistically significantly lower than those in control group( P〈0. 01). Conclusion Bevacizumab can inhibit growth of ovarian cancer cells with high expression of HER- 2 / neu,the mechanism may be related to regulating effects of p53 gene and HER-2 / neu gene.
