摘 要:目的观察Apelin-13对兔脑外伤自噬相关蛋白的影响,探讨其对兔脑外伤后自噬相关蛋白调节的作用机制。方法取健康雄性新西兰大白兔30只,随机分为假手术(Sham)组、生理盐水(Saline)组、Apelin-13组。Apelin-13组和Saline组在建立脑外伤模型前10min,分别注射Apelin-13(0.3μg/g体质量)或等量生理盐水于侧脑室,然后利用Western blot检测脑外伤后24、48h组大脑皮质自噬相关蛋白微管相关蛋白1轻链3(LC3)、p62、Beclin-1和B细胞淋巴瘤/白血病-2(bcl-2)等的表达。结果与Sham组比较,在脑外伤后24、48h,Saline组自噬相关蛋白LC3—Ⅱ/LC3-I比值(1.147±0.125),Beclin-1的蛋白表达(1.035±0.112),均显著上调(P〈0.01),同时bcl-2、p62的蛋白表达水平(0.613±0.087、0.608±0.085)显著下调。然而,与Saline组比较,Apelin-13组可显著下调自噬相关蛋白LC3-Ⅱ/LC3-1比值(0.226±0.048)和Beclin-1的蛋白表达水平(0.181±0.039,P〈0.01),同时显著上调bcl-2和p62的表达水平(0.385±0.062和0.360±0.035),亦明显降低Beclin-1/bcl-2的比值(0.704±0.064)。结论Apelin-13可以明显抑制脑外伤后兔模型大脑皮质细胞的自噬活性,这一作用可能是通过调节Beciin-1、bcl-2蛋白表达水平来实现的。Objective To observe the effect of Apelin - 13 on rabbit traumatic brain injury (TBI), and explore the relationship between Apelin - 13 and autophagy in TBI. Methods Thirty New Zealand white rabbits were randomly divided into sham operation group, saline group and Apelin - 13 group. Apelin - 13 and saline groups were pretreated with an injection of Apelin - 13 (0. 3 μg/g) and saline into the rabbit brain lateral ventricle respectively 10 min before TBI. The autophagy - associated proteins microtubule - associated protein 1 light chain 3 ( LC3), p62, Beclin - 1 and B cell lymphoma/leuke- mia - 2 ( bcl - 2 ) in the rabbit cerebral cortex were assessed by Western blotting 24 h and 48 h after TBI. Results As compared with Sham group, protein LC3 - Ⅱ/LC3 - Ⅰ ratio and Beclin - 1 expression were up -regulated (1. 147± 0. 125 and 1. 035± 0. 112) , while protein bcl -2 and p62 were downregulated in saline group 24 h and 48 h after TBI (0. 613 ±0. 087 and 0. 608 ±0. 085). As compared with saline group, Apelin - 13 decreased protein LC3 - Ⅱ/LC3 - Ⅰ ratio and Beclin - 1 expression (0. 226±0. 048 and 0. 181 ±0. 039), increased protein bc1-2 and 1362 expression (0. 385±0. 062 and 0. 360±0. 035), and downregulated protein Beclin -1/bcl -2 ratio after TBI (0. 704 ± 0. 064 ). Conclusion Apelin- 13 suppresses autophagy in rabbit TBI models possibly by down - regulating protein Beelin - 1/bel - 2 ratio.
