摘  要:目的:提取分离野生金樱子中的多糖并分析其抗氧化活性。方法:采用超声波辅助-热水提取湘西野生金樱子中的多糖,通过除色素、除蛋白、醇沉等步骤纯化,以抗坏血酸作参照物,考察了金樱子多糖对油脂氧化体系的抑制作用、对羟基自由基(·OH)和超氧阴离子自由基(O_2^-·)的清除能力及对三价铁离子(Fe^(3+))的还原效果。结果:对植物油和动物油的保护率为50%时的金樱子多糖质量浓度分别为2.8 ng·m L^(-1)和5.9 ng·m L^(-1),对羟基自由基的清除能力为50%时的金樱子多糖浓度为4.7 ng·m L^(-1),同浓度的金樱子多糖对超氧阴离子自由基清除率为23%,金樱子多糖能较好地还原三价铁离子。结论:金樱子多糖具有优良的抗氧化活性,并且其抗氧化活性与多糖质量浓度呈良好的剂量相关效应。Objective:To extract and separate polysaccharides from wild Rosae Laevigatae Fructus and analyze their antioxidant activity.Methods:Polysaccharides were extracted by ultrasonic-assisted hot water as liquid from wild Rosae Laevigatae Fructus in western Hunan Province and purified through depigmentation,deproteinization and alcohol-precipitation.With the same concentration of ascorbic acid as the control,polysaccharides were investigated for their inhibition effect on oxidation system of oils and fats,scavenging effect on ·OH and O-2,and deacidize effect on Fe-(3+).Results:When the protection rates were 50% for vegetable and animal oils,the mass concentrations of Rosae Laevigatae Fructus polysaccharides were 2.8 ng·m L-(-1) and 5.9 ng·m L-(-1),respectively.The scavenging rate was 50% for·OH,while only 23% for O_2--·at the same mass concentration of 4.7 ng·m L-(-1) polysaccharide.The polysaccharides extract exhibited strong ability of reduction to Fe-(3+).Conclusion:The polysaccharide extract shows good antioxidant ability.Further more,the antioxidant ability is well dose-dependent on the concentration of Rosae laevigatae polysaccharides.
