摘  要:目的:观察一种新型双孔钾离子通道亚单元TASK-1在大鼠海马CA1区的表达及其在局灶脑缺血再灌注后的动态变化,探讨TASK-1表达与星形胶质细胞活化的关系。方法:建立大鼠大脑中动脉阻塞(MCAO)再灌注模型,采用免疫荧光组化和激光扫描共聚焦观察大鼠MCAO再灌注后3、7、28 d海马CA1区TASK-1和GFAP表达。结果:成年大鼠海马CA1区可见大量TASK-1和GFAP双标阳性的细胞,MCAO再灌注后3、7、28 d海马CA1区TASK-1和GFAP表达逐步上调,与假手术组比较均有显著差异(P<0.05)。结论:脑缺血再灌注后大鼠海马CA1区TASK-1的表达上调与星形胶质细胞的活化有一定关系。Objective: To investigate changes in TASK-1 expression and its involvement in astrogliosis following cerebral ischemia in rat CA1 region of hippocampus. Methods: Focal ischemia was induced by temporary middle cerebral artery occlusion( MCAO). The experimental rats induced by MCAO were sacrificed on days 3,7 and 28 after reperfusion. The expressions of TASK-1 and GFAP on astrocytes in the CA1 region of Hippocampus were detected by immunofluorescence and confocal microscope. Results: TASK-1 was broadly expressed on astrocytes in the CA1 region of hippocampus. After MCAO,the expressions of both TASK-1 and GFAP on astrocytes were significantly increased at days 3,7,28 following reperfusion. Conclusion: After MCAO,the increased TASK-1 expression on astrocytes in rat CA1 region of hippocampus is related with astrocyte activation.
