5-LOXsiRNA inhibiting hepatocellular carcinoma HepG-2 cells in nude mice transplantation tumor growth and induce cell apoptosis

摘  要:目的探讨5-脂氧合酶(5-LOX)siRNA转染的人肝癌HepG-2细胞株裸鼠皮下移植瘤生长的影响及可能的作用机制。方法选取裸鼠18只,将其随机进行分三组:1、对照组(接种正常人肝癌HepG-2细胞株)。2、转染组(接种转染5-LOX-siRNA的人肝癌HepG-2细胞株)。3、空载体组(接种转染空质粒的人肝癌HepG-2细胞株)。观察载瘤小鼠皮下移植瘤生长及小鼠的生存情况,并于接种移植瘤3周后处死载瘤小鼠,记录各组移植瘤体积V及计算肿瘤衰退率,经Western和RTPCR检测通路信号蛋白ERK1/2、MEK1/2及caspase3、bcl-2等凋亡调节因子等表达情况。结果转染组肿瘤平均体积与转染空载体组、正常对照组比较明显减小(P<0.01),Western及RT-PCR检测发现转染组caspase3表达量明显增强,ERK1/2、MEK1/2及bcl-2等表达量明显下降(P<0.01)。结论抑制5-脂氧合酶的表达可显著抑制肝癌移植瘤的生长,其机制可能与5-LOX通过MEK/ERK途径调节凋亡基因bcl-2 caspase3的表达有关。Objective 5 - lipoxygenase (5 LOX) siRNA transfection of human liver subcutaneous HepG 2 cell lines nude mice transplantation tumor growth effects and possible mechanism of action. Methods Select at least 18, the random is divided into three groups: 1, the normal control group ( subcutaneous inoculation of normal liver HepG 2 cell lines). 2, the transfection group ( subcutaneous inoculation by 5 LOX - siRNA transfection of human liver HepG 2 cell lines). 3, transfection empty vector group ( subcutaneous immunization blank transfection vector plasmid human liver HepG 2 cell lines). Observe its parent subcutaneous transplanta- tion tumor growth in mice and the survival of mice, and executed after the inoculation of transplantation tumor 3 weeks tumor mice, record each transplanted tumors had V and calculation rate of tumor recession, the Western and rt-pcr detection channel signal protein ERK1/2, MEK1/2 and caspase3, BCL - 2 and apoptosis regulation factor expression. Results Transfection group average tumor vol- ume compared with transfection empty carrier group and normal control group significantly decreased (P 〈 0. 01 ), Western and rt-pcr detection caspase3 expression quantity found transfection group was obviously enhanced, ERK1/2, MEK1/2 and the BCL - 2 expression quantity decreased obviously ( P 〈 0. 01 ). Conclusion Inhibit the expression of 5 - lipoxygenase can significantly inhibit the growth of transplanted tumor of liver cancer, its mechanism may be related to 5 - LOX through the MEK/ERK pathway regulating apoptosis related gene expression of BCL - 2 caspase3.
