摘 要:目的:检测子宫内膜癌组织标本中Pak4及其活性形式p-Pak4^(ser474)的表达,分析其与临床病理参数的关系。方法:免疫组化法检测子宫内膜组织中Pak4及pPak4^(ser474)表达,应用免疫组化评分半定量分析Pak4表达与临床病理参数的关系。结果:正常子宫内膜组织、不典型增生内膜、子宫内膜癌和淋巴结转移灶中Pak4表达分别为4.00±2.88、7.20±1.10、9.69±2.83和12.00±0.00,p-Pak4^(ser474)表达分别为2.87±2.28、6.80±1.10、8.81±3.04及11.60±1.27,差异均有统计学意义(P<0.001)。子宫内膜癌组织中Pak4及p-Pak4^(ser474)表达与FIGO分期(P=0.003,0.001)、细胞分化(P=0.001,<0.001)、肌层侵犯(P=0.029,0.009)及脉管浸润(P=0.046,0.005)呈正相关。结论:Pak4及p-Pak4^(ser474)在内膜癌组织中高表达,Pak4可能促进内膜癌的恶性进展。Objective: To assess the expressions of Pak4 and phosphorylated Pak4( pPak4^ser474) in endometrial cancer tissues and correlated with clinicopathological parameters.Methods: The expressions of Pak4 and p-Pak4^ser474 were examined by immunohistochemistry.We calculated a composite histoscore to account for both stain intensity and uniformity.The expressions of Pak4 and p-Pak4^ser474 in endometrial cancer tissues were assessed and correlated with clinicopathological parameters. Results: Pak4 and activated Pak4 expression were significantly increased in atypical endometrial hyperplasia( histoscore = 7.20 ± 1.10,6.80 ± 1.10) and invasive cancers( histoscore = 9. 69 ± 2. 83,8. 81 ± 3. 04) compared with normal endometrium( histoscore = 4.00±2.88,2.87±2.28).Both were even higher in lymph node metastases( histoscore = 12.00±0.00,11. 60 ± 1. 27). We found a significantly positive correlation between Pak4 and p-Pak4 expression with disease stage( P = 0. 003,0. 001),poor histological differentiation( P = 0.001,0.001),depth of myometrial invasion( P = 0.029,0.009),vascular space invasion( P = 0.046,0.005) in endometrial cancer.Conclusion: Pak4 expression is specifically increased in the most advanced lesions,suggesting that overexpression and / or activation of Pak4 might contribute to malignant progression of human endometrial cancer.
