摘 要:目的探讨低氧和高CO_2环境下体外大鼠脑微血管内皮细胞的活性变化。方法选择15只Wistar大鼠(80~100 g),采用两步酶消化法提取大鼠脑微血管内皮细胞(brain microvascular endothelial cell,BMEC)分为3组:对照组(O_2浓度为21%);低氧组(O_2浓度为1%);高CO_2组(1%O2+15%CO_2+84%N2)。对提取的BMEC进行鉴定,检测细胞液在低氧/高CO_2环境下的血气变化,检测BMEC的活力变化。结果缺氧后细胞培养液酸碱度(pH)、氧分压(PO_2)逐渐降低,显著低于正常氧细胞培养液的pH和PO_2(P〈0.05)。高CO_2组PCO_2显著高于常氧组和缺氧组(P〈0.05)。与对照组相比,低氧导致脑内皮细胞生长活性下降,高CO_2组的内皮细胞生长状态有所改善(P〈0.05)。结论高CO_2可以减轻低氧所导致的内皮细胞损伤,促进BMEC的生长。Objective To investigate the changes of brain microvascular endothelial cells in rats with hypoxia and high CO_2 environment. Methods Fifteen Wistar rats( 80 ~ 100 g) were selected,and the brain microvascular endothelial cells were extracted by the two step enzyme digestion method,and were divided into 3 groups: control group( O_2= 21%),hypoxia group( O_2= 1%),high CO_2group( 1% O2+ 15% CO_2+ 84% N_2). The BMEC was identified,and the blood gas changes of the cells and the PO_2,PCO_2 and p H values change were detected in hypoxia and hypercapnia environment,and the activity of BMEC was detected. Results the pH and the oxygen partial pressure( PO_2) of the cells decreased gradually. p H and PO_2 were significantly lower than those of the normal oxygen cell culture. The PCO_2 in the high CO_2 group was significantly higher than that in the normal oxygen group and the hypoxia group( P〈0. 05). Compared with the control group,the growth activity of brain endothelial cells decreased,and the growth state of the cells in the high CO_2 group was improved( P〈0. 05).Conclusion CO_2 can reduce the endothelial cell damage caused by hypoxia,and promote the growth of BMEC.
