摘 要:【目的】探讨小鼠和食蟹猴帕金森病(PD)模型中肝脏变化。【方法】选用雄性C57BL小鼠20只,16月龄,随机分为正常组和造模组,每组10只,造模组采用腹腔注射1-甲基-4-苯基1,2,3,6-四氢吡啶(MPTP,剂量为25 mg/kg)5周。雄性食蟹猴8只,10岁左右,随机分为正常组3只及造模组5只,造模组以MPTP静脉注射30 d(剂量为0.2 mg·kg-1·d-1)。动物造模过程中观察行为学并记录。食蟹猴造模成功后,抽血测量肝功能。动物麻醉安乐死后,取肝脏,进行苏木精—伊红(HE)、糖原(PAS)染色,并观察病理学改变。【结果】PD小鼠模型和食蟹猴模型成功建立,食蟹猴肝功能检测结果显示:造模组总蛋白和白蛋白含量均显著降低(P<0.05)。PD小鼠肝索细胞排列紊乱,肝窦扩张,肝细胞变性坏死,糖原减少。PD食蟹猴肝细胞排列相对紊乱,肝窦明显扩张,糖原明显减少。肝细胞发生变性、坏死,肝细胞核深染,肝细胞间充满粉红色的、粗细不等的颗粒状物,有程度不等的炎性细胞浸润。【结论】帕金森病的发病可能与肝脏功能和结构改变相关。Objective To investigate the histology and function changes of liver in mouse and cynomolgus monkey models of Parkinson's disease(PD). Methods Twenty male C57 BL mice were randomly divided into model group and normal control group,10 mice in each group. Five-week intraperitoneal injection of 25 mg/kg of 1-methyl-4-phenyl-1、 2、 3、 6-tetrahy dropyri-dine(MPTP) was used for the establishment of mice model of PD. Eight male cynomolgus monkeys aged about 10 years old were also randomly divided into model group and normal control group, and the 5 monkeys of the model group were given intravenous injection of MPTP(0.2 mg·kg^-1·d^-1)for 30 days to induce PD. All of the normal control groups were injected with the same volume of normal saline. The physical behavior of the two kinds of animal models was monitored during the experiment and their hepatic histology was examined after hematoxylin-eosin(HE) staining and periodic acidSchiff(PAS)staining at the end of the experiment. And hepatic function of the cynomolgus monkey was examined after the establishment of PD model. Results The two kinds of animal models have been successfully established.In cynomolgus monkey model,total protein content and albumin content were decreased as compar ed with the normal control group(P〈0.05). PD mice had disorganized hepatocytes,expand hepatic sinusoid,degenerative and necrotic liver cells and decreased glycogen particles. The same histopathological disorders were also found in PD cynomolgus monkeys. Moreover,PD monkeys had heavily-labeled hepatocyte nuclei,fully filling with pink and unevenly granule-like substance among the hepatocytes,and various degrees of inflammatory cell infiltration.Conclusion The pathogenesis of PD may be associated with the changes of liver function and histopathology.
