摘 要:目的探讨microRNA-27b(miR-27b)对口腔鳞状细胞癌细胞增殖的影响及其作用的分子生物学机制。方法 MTT方法检测细胞增殖水平;实时定量PCR方法检测miR-27b表达水平以及卷曲蛋白7(Frizzled7,FZD7)、cyclin D1和c-myc的信使RNA(mRNA)表达水平;Western blot方法检测FZD7的蛋白表达水平;荧光素酶报告基因方法检测miR-27b与FZD7的靶向关系以及Wnt信号通路的活性。结果 miR-27b在口腔鳞状细胞癌细胞株中表达降低;过表达miR-27b可以显著抑制口腔鳞状细胞癌细胞的增殖;miR-27b可以靶向FZD7并抑制FZD7的表达及其下游的Wnt信号通路。结论 miR-27b通过靶向抑制FZD7的表达来抑制口腔鳞状细胞癌细胞的增殖,其机制可能与抑制FZD7介导的Wnt信号通路的激活有关。Objective To investigate the role of microRNA-27b( miR-27b) in regulating the cell proliferation of oral squamous cell carcinoma and the underlying molecular mechanism. Methods Cell proliferation was measured by MTT assay. The expression of miR-27 b,the mRNA expression levels of Frizzled7( FZD7),cyclin D1 and c-myc were detected by real-time quantitative polymerase chain reaction( q PCR). The protein expression of FZD7 was detected by Western blot analysis. The targeted relationship between miR-27 b and FZD7,and the activity of Wnt signaling pathway were examined by luciferase reporter assay. Results The expression of miR-27 b in oral squamous cell carcinoma cell lines was significantly decreased. Overexpression of miR-27 b markedly inhibited the cell proliferation of oral squamous cell carcinoma. miR-27 b could target and inhibit FZD7 expression and suppress Wnt signaling pathway. Conclusions miR-27 b could inhibit the cell proliferation of oral squamous cell carcinoma through targeting and inhibiting FZD7 and FZD7-mediated Wnt signaling pathway.
