摘 要:目的探讨3,3-二吲哚基甲烷(3,3-Diindolylmethane,DIM)对胃癌细胞SGC-7901增殖和侵袭的抑制作用。方法用MTT法检测不同浓度(0、10、20、40、80μmol/L)DIM对胃癌细胞SGC-7901生长抑制情况;Transwell小室检测40μmol/L的DIM对胃癌SGC-7901细胞侵袭力的影响,Western blot检测胃癌细胞中血小板源性生长因子-D(PDGF-D)和β-catenin蛋白表达的变化,ELISA法检测培养液上清中MMP-2、MMP-9的浓度。结果不同浓度的DIM对胃癌细胞SGC-7901均有抑制作用(P<0.05),随着药物浓度增加,抑制作用增强;但40μmol/L和80μmol/L差异无统计学意义(P>0.05)。40μmol/L的DIM作用24 h后胃癌细胞SGC-7901侵袭力下降75.2%(125.4±10.8 vs 31.1±4.5,P<0.05),PDGF-D和β-catenin的表达分别下降了55.8%和52.5%(P<0.05),培养液上清中的MMP-2、MMP-9的浓度明显下降(23.5±2.6 vs 13.2±1.9;30.4±2.4 vs 15.8±2.1,P<0.05)。结论 DIM对胃癌细胞的增殖和侵袭有抑制作用,其机制可能与PDGF-D、β-catenin、MMP-2、MMP-9的表达下降有关。Objective To determine the effect of 3,3-diindolylmethane (DIM) on inhibiting proliferation and invasion in gastric carcinoma SGC-7901 cells. Methods The proliferation of gastric cancer SGC-7901 cells treated with different concentrations (0, 10, 20, 40 and 80 μmol/L) of DIM was detected by MTT assay. The invasive ability of SGC-7901 cells treated with 40 μmol/L of DIM was detected with Transwell chamber assay. The expression of PDGF-D and β-catenin in SGC-7901 cells and the concentrations of MMP-2 and MMP-9 in culture supernatant were detected by Western blotting and ELISA, respectively. Results Different concentrations of DIM inhibited the proliferation of gastric cancer cells (P〈0.05), and the inhibitory effect was associated with DIM concentration. There was no statistically significant difference between 40 and 80 μmol/L. The invasive ability of SGC-7901 cells was significantly reduced by 75.2% after treatment with 40 μmol/L of DIM for 24 h (125.4±10.8 vs 31.1±4.5, P〈0.05). The expression of PDGF-D and β-catenin was decreased by 55.8% and 52.5%, respectively (P〈0.05). The concentrations of MMP-2 and MMP-9 in culture supernatant were significantly decreased after DIM treatment (23.5±2.6 vs 13.2±1.9; 30.4±2.4 vs 15.8±2.1, P〈0.05). Conclusion DIM inhibits gastric carcinoma cell proliferation and invasion through down-regulating the expression of PDGF-D, β-catenin, MMP-2 and MMP-9.
