摘 要:目的应用Luminex技术筛选结直肠癌外周血微小RNA(miRNA,miR)指纹图谱。方法收集70例结直肠癌患者、60例对照健康人和20例胃癌患者外周血血清,提取RNA,应用Luminex技术筛选结直肠癌外周血5种miRNA指纹图谱,并分析结直肠癌特征性miRNA表达与患者临床病理特征和预后的关系。结果筛选出结直肠癌特征性miRNA2种,分别为miR-29a和miR-92a,两者在结直肠癌中的表达高度一致,术后血清中的表达显著降低(二者术前术后相对表达量中位数分别为miR-29a:3.01和2.05,miR-92a:4.09和2.01,z值分别为-5.77、-3.51、-3.30,P〈0.01),且均与结直肠癌患者肿瘤细胞分化程度、血管浸润、浸润深度、淋巴结转移和临床分期明显相关(∥值分别为37.686、37.686、7.504、40.519、43.636,P〈0.05),miR-29a和miR-92a高表达的患者预后生存时间中位生存时间均为41个月,显著短于正常表达的患者(P〈0.05)。结论结直肠癌特征性miRNA指纹图谱能够反映结直肠癌进展和评估预后。Objective To screen the microRNA fingerprints of peripheral blood in colorectal canc- er patients by Luminex technique. Methods Peripheral blood serum samples of 70 eolorectal cancer patients and 20 gastric cancer patients as well as 60 contrast samples were collected to extract RNA, the Luminex technique was used to screen 5 kinds of microRNA fingerprints, and the relationship between spe- cial miRNA of colorectal cancer and the clinicopathological features and prognosis of patients was analyzed. Results Two kinds of miRNA in colorectal cancer patients were screened, respectively miR - 29a and miR-92a. The relative expression level of 2 miRNAs were decreased significantly after operation( the media value of two miRNAs before and after operation: miR -29a: 3.01 and 2.05;miR -92a:4. 09 and 2. 01 ,all P 〈0. 01 ). Their expression levels in colorectal cancer were highly consistent and both were sig- nificantly correlated with tumor differentiation, tumor vascular stages (X2 values : 37. 686, 37. 686, 7. 504, 40. 519, 43.636 ,P 〈 0. 05 ). High miR- 29a and miR- 92a expression patients had significantly shorter survival time (the media survival time of high expression of two miRNA was 41 months) than the normal expression patients ( P 〈 0.05 ). ( P 〈 0.05 ). Conclusion The specific miRNA diagnostic fingerprints of colorectal cancer can reflect the occurrence, development and prognosis assessment of colorectal cancer.
