摘 要:目的观察慢传输型便秘型(STC)患者与正常人结肠组织蛋白质表达的差异。方法采用双向凝胶电泳(2-DE)技术,计算机辅助的图像分析以及质谱鉴定方法,对5例STC患者与5例正常人结肠组织蛋白质进行分离和比较分析。结果正常对照组平均胶蛋白斑点数为239个,STC组平均胶蛋白斑点数为215个,与正常对照组平均胶匹配点数为197个,匹配率为82%,有14个蛋白质点的表达量差异有统计学意义(差异倍数〉10,P〈0.05),其中有12个蛋白点表达发生明显上调,有2个蛋白点表达发生明显下调。结论STC患者与正常人结肠组织蛋白质表达谱差异有统计学意义,这些差异蛋白可能参与了STC的发病机制.Objective To investigate and screen the different expression patterns of proteins be- tween slow transit constipation (STC) group and normal group using a comparative proteomic approach. Methods Two - dimensional eleetrophoresis was applied to separate the proteins in specimens from 5 STC patients and 5 normal controls. The proteins with statistically significant differential expression between two groups were identified by using computer aided image analysis and matrix assisted laser desorption ioniza- tion tandem time of flight mass spectrometry (MALDI- TOF- MS). Results A total of 239 protein spots were identified in the average gel of the normal controls, and 215 in patients with STC. A total of 197 pro- tein spots were matched and the mean matching rate was 82%. There were 14 protein spots which were ex- pressed with statistically significant differences. Of those 14 protein spots, the expression of 12 spots was increased markedly, while 2 were decreased significantly. Conclusion The proteomics expression in co- lonic specimens of STC patients is statistically significantly different from that of normal contrils, which may be associated with the pathogenesis of STC.
