摘  要:目的探讨白藜芦醇对肝癌的体内抑瘤作用及其机制。方法裸鼠皮下注射H22肝癌细胞建立移植瘤模型,随机分为对照组[生理盐水10ml/kg,灌胃(ig)]、白藜芦醇高剂量组(20mg/kg,ig)、低剂量组(10mg/kg,ig)和5-氟尿嘧啶(5.Fu)组[10mg/kg,腹腔注射(ip)],连续给药10d,游标卡尺测量裸鼠体内肿瘤大小,绘制肿瘤生长曲线,处死裸鼠后称量瘤重并计算抑瘤率。采用实时定量聚合酶链反应(Real-timePCR)检测瘤组织中真核翻译延长因子1A1(eEF1A1)、第10号染色体磷酸酶和张力蛋白同源丢失基因(PTEN)、半胱氨酸天冬氨酸特异性蛋白酶(Caspase)-3和Caspase-9mRNA表达水平,采用Westernblot检测eEF1A1的蛋白表达以及磷脂酰肌醇激酶(P13K)p85、蛋白激酶B(Akt)和雷帕霉素靶蛋白(mTOR)的蛋白磷酸化水平。结果白藜芦醇可显著抑制裸鼠体内肿瘤的生长,高、低剂量组的抑瘤率分别为46.1%和25.8%。Real—timePCR结果显示,白藜芦醇组瘤组织中eEFlAl的mRNA表达水平(10ms/kg:86.2±24.9;20ms/kg:63.7±14.2)较对照组(100.0±25.6)显著下降(P〈0.05),PTEN(10ms/kg:129.3±33.0;20mg/kg:174.7±31.9)、Caspase-3(10mg/kg:139.6±27.2;20mg/kg:168.8±32.3)和Caspase-9(10mg/kg:121.2±37.3;20mg/kg:184.6±46.2)的mRNA表达较对照组(PTEN:100.0±33.1;Caspase-3:100.0±29.6;Caspase-9:100.0±22.7)显著增加(P〈0.05)。Westernblot结果显示白藜芦醇组瘤组织中Akt、P13Kp85和mTOR的总蛋白表达水平无明显变化,但磷酸化水平(p-Akt:10mg/kg:89.3±19.8;20mg/kg:72.3±18.3;p-p85:10mS/kg:91.3±12.9;20mg/kg:81.2±13.2;p-mTOR:20ms/kg:64.24-13.7)均较对照组(p-Akt:100.0±27.1;p-p85:100.0±22.5;p-mTOR:100.0±27.4)显著下降(P〈0.05)�Objective To explore the anti -tumor effect and mechanism of resveratrol (Res) on H22 hepatocellular carcinoma in BALB/c nude mice. Methods The subcutaneously transplanted tumor models of H22 cells in nude mice were established and were randomly divided into four groups, including control ~Saline, intragastric administration (ig)], Res (10 and 20 mg/kg, ig) and 5- Fuorouracil [5 -Fu, 10 mg/kg, intraperitoneal injection (ip)] group. All mice were treatment for consecutively 10 days. The size of transplanted tumor was measured to draw the tumor growth curves. After all mice were sacrificed, the weights of tumor were weighted and the inhibitory rate was calculated. The real - time quan- titative polymerase chain reaction (Real- time PCR) were used to detect the mRNA expression levels of eukaryotic elongation factor 1 A1 (eEF1A1), phosphate and tension homology deleted on chromsome ten ( PTEN), cysteinyl aspartate specific proteinase (Caspase) - 3 and Caspase - 9 in tumor tissue. Western blotting were used to detect the protein expression levels of eEF1AI and the phosphporylation levels of phosphatidylinositol 3 kinase (PI3K) p85, protein kinase B (Akt) and mammalian target of rapamycin (mTOR) in tumor tissue. Results Res had obvious inhibitory effects on tumor growth compared with the control group, and the tumor inhibition rates in high - dose and low - dose group were 46.1% and 25.8% , respectively. The PCR results indicated Res significantly down -regulated the mRNA expression of eEFI A I (Control: 100. 0 ± 25.6; Res: 10 mg/kg: 86. 2 ± 24. 9; 20 mg/kg: 63.7 ± 14. 2), and up -reg- ulated the PTEN ( Control : 100.0 ± 33.1 ; Res : 10 mg/kg : 129.3 ± 33.0 ; 20 mg/kg : 174. 7 ± 31.9 ), Caspase-3 (Control: 100.0±29.6; Res: 10 mg/kg: 139.6 ±27.2; 20 mg/kg: 168.8 ±32.3) and Caspase -9 (Control: 100. 0 ±22.7 ; Res: 10 mg/kg: 121.2 ±37.3 ; 20 mg/kg: 184. 6 ±46.2) mRNA expression in tumor tissure( P 〈 0. 05 ).
