摘  要:目的观察长春新碱(VCR)对人急性淋巴细胞性白血病MOLT-4细胞促凋亡作用,并探讨其作用机制。方法分别用0、0.01、0.02、0.04、0.08μmol/L长春新碱处理MOLT-4细胞,然后用CCK-8法检测细胞活性,用Hochest33258对细胞核进行染色,用Rhodamine123对细胞进行染色,检测线粒体膜电位情况。用caspase 8、9、3抑制剂预处理MOLT-4细胞,检测caspase8、9、3对细胞活性的影响。结果长春新碱对细胞活性具有抑制作用,且呈明确的剂量-效应关系;细胞核染结果显示,长春新碱可以促进细胞凋亡,剂量越大,凋亡现像越明显;同时,长春新碱可以引起细胞线粒体膜电位下降,并发现caspase 9和3参与了细胞凋亡过程。结论长春新碱可以促进MOLT-4细胞凋亡,作用机制可能是线粒体膜电位下降,进而诱导caspase 9和3活化而促发细胞的凋亡。Objective To explore the apoptosis inducing effect of vincristine(VCR) on human acute lymphoblastic leukemia MOLT-4 cells and its mechanisms. Methods MOLT-4 cells were treated with 0, 0.01, 0.02, 0.04, and 0.08 μmol/L vincristine,respectively. The cell viability was detected by CCK-8 assay. The nuclei were stained by Hochest33258, and the cells were stained by Rhodamine123. The mitochondrial membrane potential was detected. Caspase 8, 9, and 3 depressant were used to pretreat MOLT-4cells, and their effects on cytoactive were detected. Results Vincristine could inhibit the cytoactive and presented definite dose-effect relationship. The nuclei staining indicated that vincristine could promote the apoptosis. The larger the dose was, the more obvious the apoptosis presented. At the same time, vincristine could induce the decrease of mitochondrial membrane potential. In addition, it was discovered caspase 9 and 3 joined the process of apoptosis. Conclusion Vincristine can promote the MOLT-4 cell apoptosis, which may be mediated by mitochondrial membrane potential decrease that further induce the caspase 9 and 3 activation and cell apoptosis.
