摘 要:研究并优化了白蔹中没食子酸含量测定时供试品溶液的最佳制备方法。采用法测定白蔹中没食子酸含量,并用正交试验法考察了提取方法、提取溶剂、溶剂用量对没食子酸含量测定结果的影响。得出最佳提取条件为:以50%(质量分数)甲醇为提取溶剂,加入药材10倍体积量的溶剂,超声提取1 h。其中提取方法因素的影响最为显著。该方法下没食子酸的提取率高,共存组分干扰小,操作简便易行,可作为白蔹及含白蔹化妆品中没食子酸含量测定时的供试品溶液制备方法。The optimal preparing method for sample of Ampelopsis Japonica(Thunb) Makino to be used for determining the content of gallic acid in it was investigated. The RP- HPLC was used as analytical method, and the influence of extraction method, extraction solvent and solvent dosage on the test results were studied in an orthogonal experiment. The results showed that the best extraction condition were as follows: the extraction solvent was 50%(mass fraction) methanol, the proportion of material to solvent was 1∶10(volume ratio), the ultrasonic time was 1 h. The established extraction method showed higher extracting rate, less coexisting interferential components, which can be used as a method of sample preparation for determining gallic acid in Ampelopsis Japonica(Thunb) Makino or relative cosmetics.
