摘 要:通过单因素和响应曲面法设计实验,研究回流温度、回流时间、甲醇浓度3个响应变量以及变量之间交互作用对提取效果的影响,对提取工艺进行优化。采用福林-肖卡法,以没食子酸为对照品,在750 nm波长测定印楝树皮中多酚含量。结果表明,印楝树皮多酚溶剂回流法提取的最佳工艺条件为回流温度70℃,回流时间90 min,甲醇浓度40.4%;响应变量影响顺序为甲醇浓度>回流温度>回流时间;印楝树皮中多酚提取得率平均为10.83%,与模型预测值相符。响应曲面法优选的印楝树皮多酚提取工艺条件可靠,提取得率稳定。The extraction conditions of polyphenol from neem bark were optimized by solvent refluxing method. The experiment was designed by single factor experiment and response surface method. The influence and the interaction of three response variables such as reflux tempera- ture, reflux time, methanol concentration were investigated. The extraction technology was optimized. The detection method of polyphenol was Folin Ciocalteu using gallic acid as standard. The determination wavelength of polyphenol content was at 750 nm. The optimum extrao- tion conditions of polyphenol in the neem bark using solvent refluxing method were reflux temperature 70 ℃, reflux time 90 min, methanol concentration 40.4 %. The influence order of response variables was methanol concentration, reflux temperature, reflux time. The average extract ratio of neem bark polyphenol was 10.83 %. The extraction conditions optimized by response surface method are reliable. The yield of neem bark pelyphenol is stable.
