摘  要:目的:检测腺苷对乳腺肿瘤细胞增殖及迁移能力的影响,并探究其潜在的分子机制。方法:以MCF-7细胞为模型,采用梯度浓度的腺苷处理细胞,MTT及细胞计数法检测腺苷对细胞增殖的影响,流式细胞术检测腺苷对细胞凋亡及周期的影响,划痕修复实验检测腺苷对细胞迁移能力的影响。结果:腺苷可显著抑制MCF-7细胞增殖,且抑制效果与腺苷浓度及作用时间呈相关性。流式细胞术检测结果表明腺苷可诱导MCF-7细胞凋亡,并引起细胞的G_2/M期阻滞。划痕实验结果表明腺苷抑制MCF-7细胞的迁移能力。实时定量PCR检测结果发现,促细胞凋亡分子Bax及Bak表达量显著升高,而凋亡抑制分子Bcl-2表达量显著下降,同时Caspase-3的表达量也显著升高。结论:腺苷可诱导MCF-7细胞凋亡,抑制细胞的增殖,同时腺苷可以抑制MCF-7细胞的迁移能力。腺苷诱导MCF-7细胞凋亡与其激活线粒体凋亡通路密切相关。Objective:To evaluate the effect of adenosine on breast tumor cell proliferation and migration, and explore the potential molecular mechanisms. Methods:The effect of adenosine on MCF -7 cells proliferation was detected by MTT and cell counting assay. Cell apoptosis and cell cycle was examined using FACS calibur flow cytometer. And the effect of extracellular adenosine on migration of MCF -7 cells was examined by scratch assay. Results:Adenosine significantly suppressed MCF -7 cells proliferation in a dose - and time - dependent manner. The flow cytometry analysis showed that adenosine could also induce breast cancer cell apoptosis. Moreover', adenosine treatment also decreased the migration of MCF - 7 cells. Real - time quantitative PCR results showed that pro - apoptotic molecules were up - regulated while anti - apoptotic molecule was decreased in response to adenosine treatment. Conclusion : Adenosine can suppress breast cancer cells proliferation, migration and induction of apoptosis via mitochondrial apoptosis pathway.
