不同剂量2-甲氧基雌二醇对恶性黑素瘤细胞中增殖、凋亡、血管新生基因含量的调节作用

摘  要:目的:研究不同剂量2-甲氧基雌二醇对恶性黑素瘤细胞的体外生长抑制作用。方法:培养黑素瘤细胞株B16,加入0μmol/L、10μmol/L、20μmol/L、30μmol/L、40μmol/L的2-ME处理后,采用MTS试剂盒测定细胞活力,Elisa试剂盒测定细胞中及培养基中增殖基因、凋亡基因以及血管新生基因的蛋白含量。结果:2-ME能够以剂量依赖性和时间依赖性的方式降低细胞活力;40μmol/L 2-ME处理后,细胞中Mcl-1、Cyr61的含量显著减低,Fas、Caspase14的含量显著升高,细胞中和培养基中HIF-1α、VEGF、SDF-1、CXCR4的含量显著减低。结论:不同剂量2-甲氧基雌二醇均能够抑制恶性黑素瘤细胞的体外生长,降低细胞活力、阻碍细胞增殖和血管新生。Objective:To study the inhibitory effect of different doses of 2-methoxy-estradiol on the growth of malignant melanoma cells in vitro.Methods:Firstly,melanoma B16 cells were cultured,and then 0μmol/L,10μmol/L,20μmol/L,30μmol/L and 40μmol/L of 2-ME were added.Last,cell viability was detected by MTS kit,and the contents of proliferation gene,apoptosis gene and angiogenesis gene in both cells and culture medium were determined by ELISA.Results:2-ME reduced cell viability in a dose-dependent and time-dependent way.After 40μmol/L of 2-ME treatment,Mcl-1and CYR61 contents in cells decreased significantly,while Fas and Caspase14 contents increased significantly.HIF-1,VEGF,SDF-1and CXCR4 decreased significantly in both cells and culture medium.Conclusions:Different doses of 2-ME can inhibit the growth of malignant melanoma cells in vitro by reducing the cell viability and inhibiting cell proliferation and angiogenesis.
