摘  要:目的发酵并纯化重组人β-防御素2(HBD2),为其生物活性的检定奠定基础。方法利用罐发酵人β-防御素2工程菌,并建立重组人β-防御素2的层析纯化工艺。结果确定了工程菌优化的最佳发酵条件为高溶氧、低温诱导策略,获得了高密度发酵工艺。建立了重组人β-防御素2的特殊纯化工艺。结论完成了重组人β-防御素2的发酵和纯化工艺研究。Objective Optimizing the fermentation conditions of engineering bacteria fusion protein of HBD-2and purifying the obtained interest protein to lay a solid foundation for the assay of biological activity of HBD-2recombinant protein.Methods Engineering bacteria was ferment under the optimized condition,then collect engineering bacteria to purify and enzyme digest the target protein.Results Optimum fermentation condition of engineering bacteriahas been confirmed.The HBD2 fusion protein purification method has been established.Conclusion Interest protein has been obtained after fermentation and purification of HBD2 recombinant engineering bacteria,and recovery rate of interest protein has been enhanced.
