Effects of different doses of special beef decoction on protecting neurologic function and peripheral blood T-lymphocyte subsets in rats with cerebral ischemia

摘  要:目的探讨不同剂量特制牛肉汤对局灶性脑缺血大鼠外周血T淋巴细胞亚群免疫活性的影响。方法将48只雄性SD大鼠分成假手术组Ⅰ、假手术组Ⅱ、模型组及低、中、高剂量特制牛肉汤组各8只。模型组和各剂量特制牛肉汤组以线栓法建立永久性右侧大脑中动脉缺血模型。低、中、高剂量特制牛肉汤组于造模后分别予200、400、600 mg/(kg·d)特制牛肉汤灌胃,假手术组Ⅱ予600 mg/(kg·d)特制牛肉汤,其余两组予等量生理盐水。分别于术后2 h及7 d采用Longa评分法进行神经功能缺损评分,并检测外周血T淋巴细胞亚群(CD3^+、CD4^+、CD8^+)水平,计算CD4^+/CD8^+值;于术后7 d采用2,3,5-三苯基氯化四氮唑染色法测定脑梗死体积分数。结果术后2 h和7 d,6组神经功能缺损评分,差异均无统计学意义(P>0.05)。术后7 d,假手术组Ⅰ和假手术组Ⅱ大鼠脑组织均未见梗死灶,模型组和各剂量特制牛肉汤组均可见梗死灶,中、高剂量特制牛肉汤组的脑梗死体积分数较模型组明显降低(P<0.05)。模型组外周血CD3^+、CD4^+水平及CD4^+/CD8^+值明显低于假手术组Ⅰ(P<0.05);与模型组相比,高剂量特制牛肉汤组外周血CD3^+水平升高(P<0.05),低、中、高剂量特制牛肉汤组外周血CD4^+水平升高(P均<0.05)。两组假手术组在脑梗死体积和外周血T淋巴细胞亚群水平方面比较,差异均无统计学意义(P>0.05)。结论特制牛肉汤可改善脑缺血大鼠的神经功能,对外周免疫抑制有调节作用,其机制可能与其升高外周血CD3^+、CD4^+水平有关。Objective To explore the effects of different doses of special beef decoction(BD) on protecting the neurologic function and the levels of peripheral blood T lymphocyte subsets in rats with cerebral ischemic injury. Methods Forty-eight male SD rats were divided into sham I group,sham Ⅱ group,model group,low-dose BD group,moderate-dose BD group and high-dose BD group,with 8 rats in each group. The permanent right middle cerebral artery occlusion model was established by suture method in the model group and each BD group. After modeling,the rats in the low-,moderate- and high-dose BD groups were lavaged with BD at the dose of 200,400,600 mg/(kg · d) ,respectively, in the sham Ⅱ group were lavaged with BD of 600 mg/(kg · d) ,and in the sham I group and model group were lavaged with the same volume of normal saline. At 2 hours and 7 days after operation, the neurologic impairment was assessed with Longa Scale, the levels of peripheral blood T-lymphocyte subsets(CD3 ^+ ,CD4^ + and CD8^+) were detected by flow cytometry, and ratio of CD4 ^+/CD8 ^+ was calculated. The infarct volume fractions were measured by 2,3,5-triphenyhetrazolium chloride staining at 7 days after operation. Results At 2 hours and 7 days after operation, there was no significant difference in the neurologic impairment score among 6 groups ( P 〉 0.05 ). At 7 days after operation, infarction was not observed in the brain tissues of rats in the sham I group and sham II group, but was observed in the model group and each BD group, and moderate- and high-dose BD groups had significantly lower infarct volume fractions compared to model group ( P 〈 0.05 ). The peripheral blood CD3 ^+ and CD4 ^+ levels and CD3 ^+/CD8 ^+ ratio in the model group were significantly lower than those in the sham I group (P 〈 0.05 ).
