摘 要:采用响应曲面优化法对生地黄多糖提取中的料液比、提取温度和提取时间进行优化,确定各因素最佳的水平组合。用Sevage法脱蛋白对生地黄多糖进行纯化,并对其抗氧化活性进行研究。结果表明:在料液比1∶40(g∶m L),提取温度80℃、提取时间105 min条件下,生地黄多糖提取物中多糖的含量最高为2.84%,实际测得生地黄多糖提取产率与理论预测值相对误差较小。抗氧化活性试验表明,生地黄多糖能够保护·OH所产生的氧化损伤,对·OH有显著的清除能力,最大清除率为54.4%对应的生地黄多糖的质量浓度3.00 g/L。因此可以探索作为食品工业和制药行业的天然抗氧化剂。In this work,an extraction procedure of Rehmannia polysaccharides( RP) was established. Response surface methodology( RSM) was used to optimize the extraction parameters,such as ratio of water to raw material( A),extraction temperature( B) and extraction time( C). The optimum extraction conditions were : extraction temperature 80℃,extraction time 105 min and ratio of water to raw material 40. Under these conditions,the extraction efficiency of Rehmannia polysaccharides achieved the maximum of 2. 84%. The evaluation of antioxidant activity suggested that Rehmannia polysaccharides exhibited significant protection against hydroxyl radical induced oxidative damage,the maximum scavenging activity on OH was 54. 4%( 3. 00 mg / m L). It has a potential to be used as an antioxidant in food industry and pharmaceuticals.
