摘 要:将30日龄、体质量约4.7g的青鱼放养在12个网箱中,每箱40尾,分别投喂添加0(对照组)、0.3%、0.6%和1.2%壳寡糖的饲料50d,每种饲料3个重复,研究壳寡糖添加量对青鱼血清特定生化指标、肝和肠白介素-2(IL-2)、白介素-8(IL-8)和白介素-10(IL-10)mRNA表达量的影响。试验结果显示,与对照组比较,0.3%壳寡糖组青鱼血清乳酸脱氢酶的活性显著降低(P<0.05),0.6%壳寡糖组青鱼血清甘油三酯、葡萄糖的含量和乳酸脱氢酶的活性显著降低(P<0.05),0.3%组的肝IL-2和IL-10的表达量显著提高(P<0.05),0.6%壳寡糖组的肝IL-2和IL-8的表达量显著提高(P<0.05)。试验结果表明,在饲料中添加壳寡糖能促进青鱼幼鱼肝和肠白介素的表达,增强免疫力,降低血糖血脂含量,其中以添加0.6%壳寡糖的效果最佳。A total of 560 30 day old black carp(Mylopharyngodon piceus)juveniles with body weight of about 4.7g acclimatized for 1week were reared in 12 net cages each of 1m×1m×1mat a rate of 40 fish per cage and fed 4diets containing 0(control group),0.3%,0.6% and 1.2% of chito-oligosaccharide twice a day at a rate of 3% of total fish body weight with triplication for 50 days to study the effects of chito-oligosaccharide on specific biochemical indices in the serum and the expression of IL-2,IL-8and IL-10 mRNA in hepatopancreas and intestine of the juvenile.The lactate dehydrogenase activity was found to be decreased significantly in the fish fed 0.3% chito-oligosaccharide(P〈0.05).The levels of triglycerides and glucose and activity of lactate dehydrogenase were found to be significantly reduced in the fish fed0.6% chito-oligosaccharide(P〈0.05).The expression of IL-2and IL-10 mRNA was found to be increased significantly in the hepatopancreas of the fish fed 0.3% chito-oligosaccaride(P〈0.05).The expression of IL-2and IL-8mRNA was found to be increased significantly in the hepatopancreas of the fish fed 0.6% chito-oligosaccharide(P〈0.05).The expression of IL-2and IL-10 mRNA was found to be increased significantly in the intestine of the fish fed 0.6%chito-oligosaccharide(P 0.05).It is concluded that chito-oligosaccharide supplementation significantly decreases blood glucose and lipid levels,and significantly promotes the expression of interleukin in the hepatopancreas and intestine of juvenile black carp.It is suggested that the optimal dietary supplementation of chito-oligosaccharide be 0.6%.
