摘  要:拟克隆cDNA-AFLP技术筛选的1个水稻高温差异表达片段,通过荧光定量PCR技术研究该基因在不同处理下的表达模式。结果表明:该片段为Os07g0620200基因,产物为水稻中的热激蛋白N端的DnaJ结构域。定量分析表明,各处理均促进了该基因的表达,推测该基因是水稻的一个胁迫相关基因。2个品种比较,黄华占中该基因的表达量始终高于双桂1号,表明黄华占比双桂1号在逆境中有更好的耐受性。Using the heat-sensitive variety/line Shuanggui No.1 and heat-resistant variey/line Huanghuazhan as the materials,one differentially expressed fragment was obtained and cloned from cDNA-AFLP analysis under high temperature at the seedlings stage.The expression of this fragment were analyzed by fluorescence quantitative PCR(qRT-PCR)under stress treatment.The results indicated that this differentially expressed fragment was Os07g0620200 by sequence analysis,which protein product was canonical N-terminal domain(DnaJ)of the heat shock proteins in rice.The expression of the gene by qRT-PCR was induced under high temperature,high salt,drought and Spm treatments.These results suggested that the Os07g0620200 gene might be a stress-related gene in rice.Between the two cultivars,the expression level of this gene in heat-tolerant variety Huanghuazhan was higher than that in heat-sensitive Shuanggui No.1,indicating that in comparsion to Shuanggui No.1,Huanghuazhan possesses a greater tolerance and better stress response under adverse circumstances and it was affected less by extreme environmental conditions.
