摘  要:目的了解2014年北京市丰台区流行的麻疹野毒株的优势基因型别及特征,为进一步的麻疹防治策略提供科学依据。方法采集2014年北京市丰台辖区内疑似麻疹暴发和散发患者咽拭子标本,使用Vero/SLAM细胞分离麻疹病毒。通过逆转录-聚合酶链反应(RT-PCR)从分离到的麻疹病毒中扩增出核蛋白(nucleoprotein,N)基因C末端676个核苷酸片段,再对扩增产物进行核苷酸序列测定和分析,并以C末端450个核苷酸片段构建基因亲缘关系树,进行核苷酸、氨基酸同源性分析。结果共分离到38株麻疹病毒,均为H1a基因型。38株麻疹病毒野毒株间的核苷酸和氨基酸同源性分别为97.7%~100%、96.6%~100%;同源性最高的为H1a参考株Chin9322,核苷酸和氨基酸同源性分别为98.4%~99.1%、96.6%~99.3%;与2013年Beijing株(KJ556889/KJ556890/KJ556891)核苷酸和氨基酸同源性分别为97.7%~99.7%、97.3%~100%。结论 2014年北京市丰台区内流行的麻疹病毒野毒株优势基因型别为H1a型。Objective To investigate genotype of wild-type measles viruses circulated in Fengtai district of 2014, and provide the scientific basis for measles control and elimination. Methods Measles viruses were isolated from the specimens collected from suspected measles outbreaks and sporadic patients. Fragment of 676 bp of the carboxyl end of nucleoprotein gene were amplified using reverse transcription-polymerase chain reaction(RT-PCR)method, and then the PCR products were directly sequenced and analyzed.Finally the phylogenetic tree was constructed based on 450 nucleotide of the carboxyl end of nucleoprotein gene, and the diversity analysis were done with MEGA version. Results A total of38 wild-type measles viruses were isolated and sequenced. The phylogenetic tree analysis based on 450 nucleotide of the carboxyl end of nucleoprotein gene showed that all of the isolates belonged to H1 a subgenotype. The sequence identity matrix of 38 isolates showed that the homology of nucleotide sequence was 97.7%-100% and that of amino acid sequence was 96.6%-100%. All of the isolates had the highest homology when compared with the reference strain Chin9322 with the homology of nucleotide and amino acid sequences of 98.4%-99.1% and 96.6%-99.3%, respectively. All of the isolates had the highest homology when compared with the reference strain Beijing in 2013(KJ556889 / KJ556890 /KJ556891) with the homology of nucleotide and amino acid sequences of 97.7%-99.7% and 97.3%-100%, respectively.Conclusions Major genotypes of wild-type measles viruses circulated in Beijing Fengtai district in 2014 were H1 a subgenotype. May be more than one circulation chain were coexist in Beijing Fengtai district.
