摘  要:目的对链霉菌CPCC 200497产生的抗菌活性次级代谢产物进行研究。方法对CPCC 200497发酵液乙酸乙酯提取物进行TLC、HPLC和MS分析,发现目标化合物,采用色谱方法分离纯化目标化合物,以高分辨质谱和NMR分析对目标化合物进行结构鉴定,并进行抗菌活性测定。结果从CPCC 200497发酵培养物中发现并鉴定主要次级代谢产物——醌霉素A和C组分,它们显示强烈的抗革兰阳性菌活性,特别是对MRSA和VRE耐药菌株的活性强于万古霉素;CPCC 200497还产生其他次级代谢产物。结论醌霉素具有显著的抗耐药菌等生物活性,有望成为抗耐药菌药物开发先导化合物;CPCC 200497产生的其他次级代谢产物有待继续挖掘。Objective To identify the secondary metabolites with antibacterial activity from Streptomyces sp. CPCC 200497. Methods TLC, reversed phase HPLC and MS were used to analyze the Et OAc extract of Streptomyces sp. CPCC 200497. Chromatographic methods were applied to separate and purify the target compounds. HRMS and NMR were employed to characterize the structure of the target compounds. Results Streptomyces sp. CPCC 200497 produced different kinds of secondary metabolites. Quinomycins A and C were identified as its principal secondary metabolites. Quinomycins A and C showed very strong in vitro activities against Gram-positive bacterial strains including MRSA and VRE, with MICs lower than vancomycin, but displayed very weak or no activity against Gram-negative test bacterial strains. Conclusion Quinomycins A and C possess significant antibacterial activitied. They may be used as lead compounds in drug development against MRSA and VRE infections.
