摘  要:目的:观察荷乳腺癌小鼠电针后其视交叉上核节律输出信号分子m PK2表达水平的变化,探讨电针对荷瘤动物自发性活动节律影响的输出信号分子调控机制。方法:将18只C57雄性小鼠随机分为空白组、模型组、电针组3组,每组6只。LD驯化10天后关闭光源使动物处于自由运行状态,DD驯化10天后开始造模,造模成功继续监测节律7天后进行电针干预。电针组在CT4针刺百会和长强穴,使用韩氏穴位神经刺激仪,频率2/15Hz,电流0.5m A,每次15分钟,每日1次,连续电针3次。空白组和模型组在相应时相点CT4用同样的方法抓取和固定。第三次电针后2小时立即断颈处死动物,采用实时荧光定量PCR法测定各组小鼠视交叉上核(SCN)内PK2mRNA表达量。结果:模型组视交叉上核PK2mRNA表达水平较空白组有降低趋势,但与空白组相比没有统计学意义(P>0.05)。电针组PK2mRNA表达水平较模型组明显升高,与模型组比较有统计学意义(P>0.05)。结论:电针对荷瘤小鼠自发性活动节律的调整作用机制可能与调控SCN节律输出信号分子PK2有关。Objective: Observing the change of m PK2 in SCN of mice bearing breast cancer after electro-acupuncture for further discuss on the output signal in SCN regulating mechanism of electro-acupuncture on the spontaneous activity rhythm of animals bearing tumors. Methods: 18 male C57mice were randomly divided into 3groups with 6in each group. Each group received LD domestication for 10 days. After that,light source was blocked so that the mice were in free movement and they received DD domestication for 10 days before model establishment started. Upon successful model establishment,the spontaneous activity rhythms of mice were monitored for 7days. After that,electro-acupuncture was administered on the electro-acupuncture group.Electro-acupuncture was administered at Changqiang( GV1) and Baihui( GV20) on CT4 with HANS Acupoint and Nerves Stimulator( HANS-200) using 2 /15 Hz and 0. 5m A for 15 minutes each time and one time a day. Animals were immediately sacrificed2 hours after the third time. The sample were extracted and detected by real-time PCR. Results: The PK2 mRNA levels in SCN of the model group were lower than the blank group but there were no significant differences( P〈0. 05) respectively. The PK2 mRNA levels in SCN of electro-acupuncture group were higher than the model group which were significantly different( P〈0. 05). Conclusion: The regulating mechanism of electro-acupuncture on the spontaneous activity rhythm of tumor-bearing mice is probably in connection with the regulation of the expression of output signal PK2 in SCN which should be proved by further experiments.
