摘　　要：目的探讨ARMc8(armadillo-repeat-containing protein 8)对肺癌细胞侵袭和迁移能力的影响。方法分别在肺癌细胞A549和H1299中干扰和转染ARMc8,Transwell和划痕实验检测ARMc8对细胞侵袭和迁移能力的影响,并使用Western Blot方法检测α-catenin、E-cadherin、MMP-7和snail侵袭转移相关因子的改变。结果干扰ARMc8可明显抑制A549细胞侵袭和迁移能力,下调MMP-7和snail表达水平,上调E-cadherin和α-catenin表达水平。而转染外源性的ARMc8,可显著增强H1299细胞的侵袭和迁移能力,同时上调MMP-7和snail表达,下调E-cadherin和α-catenin表达。结论 ARMc8过表达促进肺癌细胞的侵袭和迁移,可能与其上调MMP-7和snail,下调E-cadherin和α-catenin相关。Objective To investigate the effect of ARMc8（armadillo-repeat-containing protein 8）on the invasion and migration of lung cancer cells.Methods After transfection and interference of ARMc8 respectively in A549 and H1299 cell lines,transwell and wound-healing assay were used to detect the invasion and migration ability of the lung cancer cells,meanwhile,Western Blot detected the expression of some invasion and migration related factors,such as α-catenin,E-cadherin,MMP-7 and snail.Results Interference of ARMc8 inhibited the invasion and migration of A549 cellls,downregulated MMP-7 and snail expressions,but upregulated α-catenin and E-cadherin expressions.Furthermore,transfection of ARMc8 promoted the invasion and migration of H1299 cells,upregulated MMP-7 and snail expressions,but downregulated α-catenin and E-cadherin expressions.Conclusion The promotion of the invasion and migration of lung cancer cells by ARMc8 overexpression might be related to the upregulation of MMP-7 and snail and the downregulation of E-cadherin and α-catenin.
