槲皮素对脑胶质瘤大鼠Caspase-3和Caspase-9表达的影响

摘　　要：目的探讨槲皮素对脑胶质瘤大鼠Caspase-3和Caspase-9表达的影响。方法雌性Wistar大鼠30只,随机分为3组,即假手术组、模型组和槲皮素治疗组,每组10只。建立大鼠C6胶质瘤模型,于接种后17d处死各组大鼠,采用免疫组织化学法和Western blot法检测肿瘤组织Caspase-3和Caspase-9蛋白表达水平的变化。结果与假手术组相比,模型组大鼠脑组织Caspase-3和Caspase-9的蛋白表达的平均光密度值显著降低(<0.01);与模型组相比,槲皮素治疗组大鼠脑组织Caspase-3和Caspase-9的蛋白表达的平均光密度值显著升高(<0.01)。结论槲皮素能够上调脑胶质瘤大鼠Caspase-3和Caspase-9的表达,对脑胶质瘤有潜在的治疗作用。Objective To study the effect of quercetin on the expressions of Caspase-3 and Caspase-9 in rats with brain glioma.Methods A total of 30 female rats were randomly divided into 3 group（n =10 each group）：sham group,model group and quercetin treatment group.The rat model of C6 glioma was established,and animals were killed 17 days after tumor graft.The expressions of Caspase-3 and Caspase-9 were observed by immunohistochemistry and western blot methods.Results The mean optic density（MOD）value of positive expressions of Caspase-3 and Caspase-9 in brain tissues of the rats was lower in model group than in sham group significantly（p〈0.05）,but higher in quercetin treatment group than in model group significantly（p〈0.05）.Conclusion The upregulation of Caspase-3 and Caspase-9 expressions by quercetin indicates that quercetin might have treatment effect on brain glioma.
