摘  要:目的观察NRG1、ErbB2和脊髓星形胶质细胞在骨癌痛大鼠中的表达。方法雌性SD大鼠,随机分为2组,每组48只。Sham组(假手术组)和骨癌痛模型(CIBP,胫骨内注射Walker256乳腺癌细胞)组。于注射瘤细胞前和后6、14、21d取出L4-L6节段脊髓,利用免疫组化方法明确星形胶质细胞标记物胶质细胞纤维酸性蛋白(GFAP)在脊髓背角的分布及表达,qRT-PCR检测NRG1、ErbB2和GFAP mRNA的变化。结果 CIBP组大鼠接种瘤细胞后14天,GFAP蛋白和mRNA表达水平明显高于Sham组,并持续至接种后21d(P〈0.05),星形胶质细胞的胞体变得肥大。接种瘤细胞后6d大鼠脊髓NRG1mRNA和ErbB2 mRNA表达水平比Sham组显著增加,持续至接种瘤细胞后21d(P〈0.05)。结论髓背角NRG1、ErbB2参与了骨癌痛,可能与星形胶质细胞活化相关。Objective To study the expression of neuregulin1(NRG1)and ErbB2 and GFAP in spinal cord of rats with cancer-induced bone pain(CIBP).Methods Female SD rats were randomly divided into(n=48 for each):Sham group and CIBP group(walker256 mammary gland carcinoma cells were inoculated into the rat tibia).Immunofluenrescence was used to identify the distribution and location of astrocytes marker(GFAP).The expression of NRG1,ErbB2 and GFAP mRNA was examined by quantitative reverse transcription-polymerase chain reaction(q RT-PCR).Results Compared with sham group,the expression levels of GFAP protein and mRNA in spinal cord were significantly increased from day 14 to day 21 after inoculation(p〈0.05).Compared with sham group,the expression levels of NRG1 and ErbB2 mRNA in spinal cord were significantly increased from day 6 to day 21 from day 14 to day 21 after tumor cell inoculation(p〈0.05).Conclusion The involvement of NRG1 and ErbB2 in the CIBP might be related to activation of astrocytes.
