摘  要:目的探讨在宫颈癌中光动力学调控对miR143激活Bcl-2/Bax的信号路径的影响,为光动力学联合miR143应用于宫颈癌的治疗机制奠定基础。方法人宫颈癌He La细胞分别进行miR143干扰处理(miR143组)、光动力照射处理(PDT组)及miR143干扰联合光动力照射处理(PDT+miR143组),采用CCK8法、流式细胞术及Transwell小室侵袭模型对各组处理后He La细胞的抑制率、凋亡率及侵袭能力进行分析,同时采用荧光定量PCR检测不同处理前后各组细胞的miR143、Bcl-2和Bax的mRNA表达水平。结果 3组之间的细胞抑制率、凋亡率差异有统计学意义(P<0.05),PDT+miR143组细胞抑制率和凋亡率分别高于PDT组和miR143组,差异具有统计学意义(P<0.05),而PDT+miR143组穿膜细胞数显著高于miR143组和PDT组(P<0.05)。处理后,3组miR143和Bcl-2 mRNA表达水平较处理前均显著升高(P<0.0001),且处理后PDT+miR143组miR143和Bcl-2 mRNA表达水平显著高于PDT组和miR143组(P<0.05),而处理后3组Bax mRNA表达水平无明显变化。结论在宫颈癌中,光动力学治疗对miR143激活Bcl-2/Bax的信号路径起正向调控作用,为光动力学联合miR143治疗宫颈癌的机制奠定了基础。Objective To investigate the regulation effect of photodynamic therapy on Bcl-2/Bax signal path activated by miR143 in human cervical cancer, and lay foundation for cervical cancer treated with photodynamic therapy along with miR143. Methods Human HeLa cells received miR143 interference (miR143 group), photodynamie irradiation treatment( PDT group ) and miR143 interference combined with photodynamic irradiation(PDT + miR143 group). The rate of inhibition,apoptosis and invasion were detected by CCK8 ,flow cytometry and Transwell model, respectively. Meanwhile the expression levels of miR143, Bcl-2 and Bax before and after different treatments were detected by fluorescence quantitative PCR. Results The inhibition rates and apoptosis rates among the 3 groups were significantly different ( P 〈 0.05 ), inhibition rates and apoptosis rates of PDT + miR143 group were distinctly higher than those of PDT group and miR143 group( P 〈 0.05 ). And invasion cell number of PDT + miR143 group was significantly higher than that of miR143 group and PDT group (P 〈 0.05 ). After treatment, the expression lev- els of miR143 and Bcl-2 among the 3 groups visibly increased than before treatment( P 〈 0. 0001 ). Moreover the expression levels of miR143 and Bcl-2 were significantly higher than those of PDT group and miR143 group (P 〈 0.05 ). However the expression levels of Bax mRNA among the 3 groups showed no significant differences after treatment ( P 〉 0.05 ). Conclusion In cervical cancer, photodynamic therapy upregulated the Bcl-2/Bax signal path activated by miR143, and lay foundation for cervical cancer treated with photodynamic therapy along with miR143.
