摘  要:目的探讨趋化因子受体7(CXCR7)和缺氧诱导因子1α(HIF-1α)在结直肠癌(CRC)中的表达及临床意义。方法采用免疫组化检测96例CRC组织及其癌旁正常组织中CXCR7、HIF-1α的表达,分析其与临床病理特征、预后的关系。结果 CXCR7和HIF-1α在CRC组织中的阳性表达率均高于癌旁正常组织(P<0.01)。CXCR7与HIF-1α在CRC组织中的表达呈正相关(r=0.499,P<0.01)。CXCR7和HIF-1α表达与淋巴结转移、远处转移和TNM分期密切相关(P<0.01或P<0.05),而与性别、年龄、肿瘤位置、肿瘤大小和分化程度无关(P>0.05)。CXCR7和HIF-1α阳性表达患者的5年总生存率和无进展生存率均低于阴性表达患者(P<0.01或P<0.05)。CXCR7阳性表达和远处转移是影响CRC患者预后的独立因素(P<0.01或P<0.05)。结论 CXCR7和HIF-1α在CRC组织中高表达,可能与CRC发生、发展密切相关,有望成为其预后判断和治疗的新靶点。Objective To investigate the expressions and clinical significance of C-X-C chemokine receptor 7(CXCR7)and hypoxia inducible factor-1α(HIF-1α )in colorectal carcinoma(CRC).Methods The expressions of CXCR7 and HIF-1α were detected by immunohistochemistry in96 cases of CRC tissues and corresponding tumor-adjacent normal tissues.The correlation of the expressions with clinicopathologic characteristics and prognosis was analyzed.Results The positive expression rates of CXCR7 and HIF-1α in CRC tissues were higher than those in tumor-adjacent normal tissues(P〈0.01).There was a positive correlation between expressions of CXCR7 and HIF-1α in CRC tissues(r=0.499,P〈0.01).The expressions of CXCR7 and HIF-1α were closely correlated with lymph nodal metastasis,distant metastasis and TNM staging(P〈0.01 or P〈0.05),but were not relevant to sex,age,tumor location,tumor size and differentiation degree(P〉0.05).Compared with the patients with negative expressions of CXCR7 and HIF-1α ,the overall survival rate and progression-free survival rate in 5-year follow-up were decreased in the patients with positive expressions of CXCR7 and HIF-1α (P〈0.01 or P〈0.05).The positive expression of CXCR7 and distant metastasis were the independent predictors for the prognosis of CRC patients.Conclusion CXCR7 and HIF-1α are highly expressed in CRC tissues,which play an important role in the development and progression of CRC and may serve as the new targets for therapy and indicators for predicting the prognosis.
