肾癌组织中nucleostemin表达的研究

摘　　要：目的研究肾癌组织中干细胞因子(nucleostemin,NS)的表达。方法选取接收的50例肾癌患者,患者均经过病理证实,收集切除的肾癌标本。按Thoenes进行病理分级分为G1、G2、G3 3级,临床分期按Robson分为Ⅰ、Ⅱ、Ⅲ、Ⅳ期。通过反转录聚合酶链反应(RT-PCR)检测患者肾癌组织中NS mRNA的表达,分析其相关性及意义。另外选取10例正常肾组织的志愿者,与肾癌组织患者组进行比较分析。结果正常肾组织组的吸光度明显大于肾癌组织组,差异有统计学意义(P<0.05),在肾癌细胞不同病理分型中,透明细胞癌、嗜色细胞癌、嫌色细胞癌的吸光度值比较,差异无统计学意义(P>0.05);G1、G2、G3肾癌组织NS表达阳性率分别为45.45%、77.27%、88.23%,差异有统计学意义(P<0.05);肾癌患者在Ⅰ期、Ⅱ期、Ⅲ期、Ⅳ期中NS表达阳性率分别为57.14%、61.11%、100%、100%,差异有统计学意义(P<0.05),而且Ⅲ期、Ⅳ期的阳性率明显大于Ⅰ期、Ⅱ期;NS表达阳性率与患者的性别、年龄、肿瘤大小、以及淋巴结转移情况无关(P>0.05)。结论肾癌组织中nucleostemin的表达与肾癌细胞的分型、患者的性别、年龄、肿瘤大小、以及淋巴结转移情况无关,与肾癌病理分级、分期有关。To investigate the expression of nucleostemin（ NS） in renal carcinoma tissues. Methods Fifty patients with renal cell carcinoma who were treated in our hospital from May 2013 to October 2014 were enrolled in the study. All the patients were pathologically confirmed,including 27 females and 23 females（ age range： 26 ~ 80 yr）,the renal carcinoma specimens resected surgically were colleted and pathologically graded into G1,G2,G3 according to Thoenes,which were clinically staged into stageⅠ,Ⅱ,Ⅲ,Ⅳ according to Robson. The expression levels of NS mRNA were detected by reverse transcription-polymerase chain reaction（ RT-PCR）,and the correlation and significance were analyzed. Besides 10 healthy subjects were served as control group. Results The absorbance in normal renal tissues was significantly higher than that in renal carcinoma tissues（ P〈0. 05）. However there was no significant difference in absorbance among clear cell carcinoma,chromophilic cell carcinoma,chromophobe cell carcinoma（ P〉0. 05）. The positive expression rates of NS in renal carcinoma tissues were 45. 45%,77. 27%,88. 23%,respectively in G1,G2,G3,there were significant differences among them（ P〈0. 05）. The positive expression rates of NS in stageⅠ,Ⅱ,Ⅲ,Ⅳ renal carcinoma tissues were 57. 14%,61. 11%,100%,100%,respectively,there were significant differences among them（ P〈0. 05）. Moreover the positive expression rates of NS in Ⅲ,Ⅳ stages renal carcinoma tissues were significantly high than those in stageⅠ,stage Ⅱ renal carcinoma tissues,besides,the positive expression rates of NS were not correlated to patient＇s age and sex,tumor＇s size and lymph node metastasis（ P〉0. 05）. Conclusion The expression levels of NS in renal carcinoma tissues are not correlated to types of renal cell carcinoma,patient＇s age and sex,tumor＇s size and lymph node metastasis,however,which are related with pathological grading and clinical staging of renal carcinoma.
