摘 要:目的研究抗郁丸对慢性温和不可预见性应激(CUMS)抑郁症大鼠行为学及血清中甲状腺激素、细胞因子的影响,探讨抗郁丸治疗抑郁症的作用机理。方法采用慢性温和不可预见性应激结合孤养方法构建CUMS大鼠模型。60只雄性大鼠随机分为正常组(10只)和模型组(50只),模型组造模3周后,随机分为模型对照组、丙咪嗪组及抗郁丸低、中、高剂量组,每组10只,记录大鼠体重及糖水消耗量变化,并进行开场行为检测、强迫游泳实验及悬尾实验。给药8周后处死大鼠,腹主动脉取血,分离血清,测定大鼠血清中甲状腺激素及细胞因子含量。结果模型对照组大鼠体重增长减慢,糖水消耗量减少,行为运动得分降低,强迫游泳不动时间及悬尾不动时间延长(P<0.05),血清中四碘甲状腺激素氨酸(T4)、促甲状腺激素(TSH)含量降低(P<0.05),白介素1β(IL-1β)、肿瘤坏死因子(TNF-α)及IL-6含量升高(P<0.05),与正常组相比差异有统计学意义。抗郁丸及丙咪嗪可加快模型大鼠的体重增长,增加糖水消耗量,增加行为学得分,缩短强迫游泳不动时间及悬尾不动时间(P<0.05),并提高血清中T4及TSH含量(P<0.05),不同程度降低IL-1β、TNF-α及IL-6水平(P<0.05),与模型对照组相比差异有统计学意义。结论抗郁丸能改善CUMS大鼠抑郁症状,逆转抑郁症大鼠血清中甲状腺激素及细胞因子的含量,这可能是其发挥抗抑郁作用的重要机制之一。Objective To explore the mechanism of anti-depression pill treating CUMS depression rats through the measurement of behavior,the serum levels of thyroid hormone and cytokines. Methods The rat model of CUMS was established by chronic unpredictable mild stress(CUMS) method. A total of 60 male rats were randomly divided into 2groups,normal(10 rats) and model(50 rats) groups. After 3 weeks of CUMS in model group,the rats of model group were randomly divided into model control group,imipramine group and anti-depression pill low,middle and high dose groups;each group had 10 rats. Rats body weight and consumption of sugar water were observed. Opening Behavior,Forced Swimming Test and Tail Suspension Test were applied to observe the rat behavior. After treatment for 8 weeks,the rats were sacrificed.Thyroid hormone and cytokines levels in serum were measured. Results Compared with normal group,the model control group grew slower,the consumption of sugar waterdecreased,the scores of behavior movement reduced,the Forced Swimming fixed time and the Tail Suspension fixed time extended(P〈0.05),serum T4 and thyroid-stimulating hormone(TSH) levels reduced(P 〈0.05),the levels of interleukin 1β(IL-1β),tumor necrosis factor α(TNF-α) and IL-6were elevated(P 〈0.05). Compared withmodel control group,anti-depression pill groups and imipraminecould increase the growth rate of body weight,increased the consumption of sugar water and behavior movement scores,obviously shorten the Forced Swimming fixed time and the Tail Suspension fixed time(P 〈0.05),increase the serum T4 and TSH levels(P 〈0.05),and reduced the levels of IL-1β,TNF-α and IL-6 in different degrees(P〈0.05). Conclusion Anti-depression pill couldimprove the CUMS rats depressive symptoms,reverse the serum levels of thyroid hormone and cytokines,which may be an important mechanism of the antidepressant effect of the anti-depression pill.
