高效液相色谱法测定生化汤协定方代煎液中游离阿魏酸和总阿魏酸的含量

摘　　要：目的建立高效液相色谱法（HPLC法）测定生化汤协定方代煎液中游离阿魏酸和总阿魏酸含量的方法。方法应用HPLC法测定游离阿魏酸和总阿魏酸的含量,以Diamonsil C18柱（4.6 mm×250mm,5μm）为色谱柱,甲醇-1%醋酸溶液（30∶70）为流动相,检测波长为321 nm。结果游离阿魏酸在进样量0.212～1.060μg范围内呈良好的线性关系,回归方程为Y=4.685 5×106X-2.109 0×104（r=0.999 9）,游离阿魏酸平均加样回收率100.60%,相对标准偏差（RSD）=2.33%,总阿魏酸平均加样回收率99.43%,RSD=2.65%。结论本方法操作简单,方法可靠,结果准确,灵敏度高,重现性好。Objective To establish a method for determination of free and total ferulic acid in decoction of Shenghua prescription by high performance liquid chromatography（ HPLC）.Methods On Diamonsil C18（ 4.6mm × 250 mm,5 μm）; with methyl alcohol（ containing 1 % acetic acid） = 30∶ 70 as mobile phase and UV determination at 321 nm.Results A good linearity was obtained over the range of 0.212 ～ 1.060 μg for free ferulic acid,regression equation was Y = 4.685 5 × 106X-2.109 0 × 104（ r = 0.999 9）.The average recoveries of free and total ferulic acid components were 100.60% with relative standard deviation（ RSD） = 2.33% and 99.43% with RSD = 2.65%,respectively.Conclusion This method is simple,reliable,accuracy,with high sensitivity and better repeatability.
