摘  要:目的探讨何首乌提取物二苯乙烯苷(TSG)对APP/PS1双转基因小鼠脑组织中β淀粉样前体蛋白(APP)和早老素-1(PS1)基因的表达变化。方法 3月龄APP/PS1双转基因小鼠50只,随机分为APP/PS1对照组、石杉碱甲组和TSG高、中、低(0.3、0.1、0.033 g/kg)剂量组,另取同龄C5B7L/6J小鼠10只为正常对照组。分别给予相应药物60 d后,应用FQ-PCR法检测脑组织中APP、PS1 mRNA的表达。结果与APP/PS1对照组比较,TSG高、中、低各剂量组以及石杉碱甲组APP、PS1mRNA表达均明显下调(P<0.01或P<0.05);TSG各剂量组与石杉碱甲组比较差异无统计学意义(P>0.05)。结论二苯乙烯苷可对阿尔茨海默病起到一定防治保护作用,其机制可能与APP、PS1基因受到明显抑制有关。Objective To observe the effect of TSG which extraction from Polygonum multiflorum Thunb on gene expression of β-amyloid precursor protein( APP) and presenilin-1( PS1) in APP / PS1 double transgenic mice brain tissues. Methods Fifty three-months-old APP / PS1 double transgenic mice were randomly divided into five different groups: APP / PS1 control,huperzine A,high-,mid- and low-dose of TSG( 0. 3,0. 1,0. 033 g / kg) groups,with 10 mice in each group. Another 10 same age C5B7 L /6J mice were assigned into normal control group. After medication corresponding drugs for 60 days,expressions of APP and PS1 mRNA in brain tissue were assayed by fluorescent quantitative-PCR. Results Compared with APP / PS1 control group,the expressions of APP and PS1 mRNA in all dose of TSG group and huperzine A group were down-regulated,the differences had statistical significances( P〈0. 01 or P〈0. 05); Compared with huperzine A group,all dose of TSG had no significant difference( P〉0. 05). Conclusions TSG has certain preventive and therapeutic effect on Alzheimer's disease,which may be related to the inhibition of the expressions of APP and PS1 mRNA.
