摘  要:试验旨在研究凝结芽孢杆菌的发酵条件、发酵液的浓缩方式、载体吸附等工艺,并评价其产品性能。采用10 L全自动发酵罐发酵凝结芽孢杆菌,采用有机膜和矿物质吸附剂对发酵液进行浓缩、吸附。采用体外消化试验、体外药敏试验、耐高温试验和产品储存试验评价产品性能。结果表明:1凝结芽孢杆菌制剂的最佳工艺参数为搅拌速度200 r/min,通气量350 L/h,浓缩设备为有机陶瓷膜过滤设备,矿物质吸附剂与发酵液比例为11。2产品在85℃处理10 min存活率为71%,经胃液消化后存活率为86.8%,过肠液后存活率为69%;产品在25℃和35℃储存30 d后仍保持50%作用的存活率,储存60 d后菌种数依旧可以到达108数量级。由此可见,此产品具有耐酸、耐胆盐、耐高温和易于储存等优良特性。This paper mainly aimed to explore optimal Bacillus coagulans fermentation conditions, con-centration process of the fermentation liquid, moreover it surveyed a evaluation on the performance ofthe product. The influence of stirring speed, throughput and constant p H for cell growth and spore wasanalysed by the production test. Concentrate and adsorb the fermented liquid to get condensate bacil-lus viable pharmaceutics. Then simulating pelleting temperature, digestion test in vitro, drug sensitivetest and storage test could be used to preliminarily evaluate the production. The results showed as fol-lows: ① The proper operation condition was 200 r/min of the mechanical stirring speed, 350 L/h ofthroughput, organic ceramic membrane filtration equipment treat and mineral adsorbent adsorbed theconcentrated solution by 1:1. ② The survival rate of the strain were respectively 71% after process-ing the pelleting temperature of 85 ℃ for 10 min; the survival rate was 86.8% after processing the sim-ulated gastric fluid; the survival rate could reach to 69% after experiencing simulated intestinal fluid;Drug sensitivity tests showed that the product could be used with amoxicillin; and the survival ratewere more than 50% after processing under the temperature of 25 ℃ and 35 ℃ for 30 days, beside thequantity of the strain was 108 orders of magnitude after processing under the temperature of 25 ℃ and35 ℃ for 60 d. It is concluded that the product has excellent performance with acid and bile salt resis-tance, thermostability, easy storage.
