心肌切片的苏木精-伊红染色制作方法探讨

摘　　要：目的在H-E染色心肌切片上既能观察到心肌的组织结构变化,又能清楚的显示出心肌闰盘及横纹结构。方法对成年狗心脏采用含酸性固定液固定,按常规进行石蜡包埋切片,在苏木精染色液染色后不经盐酸乙醇分色,而直接入自来水进行蓝化,随后用伊红染色液复染,常规脱水、再透明、最后用中性树胶封片。结果心肌组织结构与心肌纤维上的横纹和纤维间的闰盘显而易见,结构清晰,色彩鲜明。结论以成年动物心脏为材料、以含酸性成分的固定液固定、苏木精染色后不经盐酸乙醇分色而直接蓝化,效果满意。Objective Structural changes of myocardium and Intercalated disc and cross striation can be observed in the HE staining heart section. Methods Heart samples gained from adult dogs were contained in acid fixed fluid,conduct conventional paraffin embedding and sliced,without color separation by hydrochloric acid ethanol after hematoxylin staining fluid dyeing,directly dropped into the tap water,and then re- stain by using eosin staining fluid,finally dehydration and trans- parent and sealing piece. Results Myocardial tissue structure and the cross striation on the myocardial fibers and the fibers of intercalated disc were obvious,the structure was clear and brightly. Conclusions Chose adult animal heart tissue as materials,fixed liquid containing acid composition,without color separation by hydrochloride ethanol after hematoxylin staining,obtained satisfactory effects.
