摘  要:目的 探讨S100A6基因过表达对于肺腺癌细胞恶性表型的影响。方法构建重组pLeno—DCE—S100A6载体,慢病毒转染A549肺腺癌细胞,应用实时荧光定量聚合酶链反应和Western blot鉴定S100A6基因和蛋白表达;采用四甲基偶氮唑蓝法、Transwell和流式细胞仪分别检测细胞增殖、侵袭、细胞周期及凋亡等恶性表型特征。结果转染S100A6基因的A549肺腺癌细胞S100A6mRNA和蛋白的表达较空载体对照组和阴性对照组均有明显的上调(P值均〈0.05);细胞增殖和侵袭能力较空载体对照组和阴性对照组增加(P值均〈0.05);细胞周期比例在G0/G1期与空载体对照组和阴性对照组比较差异无统计学意义(P值均〉0.05),在S期显著高于其他两组(P值均〈0.05),在G。/M期显著低于其他两组(P值均〈0.05);平均凋亡率较空载体对照组和阴性对照组下降(P值均〈0.05)。结论肺腺癌细胞S100A6基因过表达可增强肿瘤细胞的增殖和侵袭能力,增加DNA合成,减弱凋亡等,与肿瘤发生、发展、侵袭及转移等生物学行为密切相关,有望作为肺腺癌诊断和治疗的分子标志物。Objective To investigate the influence of S100A6 gene overexpression on the malignant phenotype of lung adenocarcinoma cell. Methods The recombination pLeno-DCE-S100A6 vector was constructed, followed by transfection in A549 lung adenocarcinoma by lentivirus. The malignant phenotypes including cell proliferation, invasion, cell cycle and apoptosis were detected by MTT, Transwell, and flow cytometer methods. Results The expressions of S100A6 mRNA and protein of A549 lung adenocarcinoma cell in pLenO-DCE-S100A6 group were obviously increased than those in empty vector group and negative control group (all P 〈0.05). The ability of proliferation and invasion in pLenO- DCE-S100A6 group was significantly increased than that in empty vector group and negative control group (all P 〈0.05). There was no statistical difference in the cell proportion of Go/GI phase between pLenODCE-S100A6 group and empty vector group or negative control group (all P 〈 0.05), while the cell proportion of S phase in pLenO-DCE-S100A6 group was significantly higher than that in the two control groups (all P 〈0.05), and the cell proportion of G2/M phase was significantly lower than that in the two control groups (all P 〈0.05). Condusions S100A6 gene involves in the progress of malignant phenotype of lung adenocarcinoma cell and has close correlation with occurrence, development and metastasis of tumor. It is hopeful to become a target spot for diagnosis and treatment of lung adenocarcinoma.
