摘 要:目的探讨环磷腺苷效应元件结合蛋白(c AMP-response element binding protein,CREB)在不同级别脑胶质瘤组织中的表达及其临床意义。方法采用免疫组化法检测CREB在5例颅脑损伤减压术后脑组织切片,55例不同分化程度脑胶质瘤组织切片中的表达,并采用Western blot、RT-PCR检测CREB在10例颅脑损伤减压术后脑组织标本,30例不同分化程度脑胶质瘤组织中的表达。结果免疫组化法测得CREB的阳性表达率分别为对照组2/5,Ⅰ-Ⅱ级胶质瘤10/15,Ⅲ级胶质瘤11/12,Ⅳ级胶质瘤28/28,免疫组化显示数据总体上有统计学差异(H=28.183,P<0.05)。RT-PCR法检测2^(-ΔΔCt)值,对照组:1.00±0.00,Ⅰ-Ⅱ级胶质瘤:1.35±0.07,Ⅲ级胶质瘤:2.88±0.11,Ⅳ级胶质瘤:3.75±0.20。脑胶质瘤组织中CREB的表达均高于对照组脑组织,且各级别癌组织间有统计学意义(F=1.208,P<0.05)。Western blot法测得对照组:0.311±0.014,Ⅰ-Ⅱ级胶质瘤:0.469±0.026,Ⅲ级胶质瘤:0.641±0.028,Ⅳ级胶质瘤:0.896±0.024,各组间有统计学差异(F=1.123,P<0.05)。结论 CREB在不同分化程度脑胶质瘤组织中均存在过表达现象,与肿瘤的分化程度呈正相关,可能在胶质瘤发生发展过程中起着重要的调节作用。Objective To investigate the clinical significance and abnormal expression of the CREB in different grade gliomas. Methods The expression of CREB was examined by using immunohistoehemistry in brain tissues from the brain injury (5 eases) and different grade gliomas (55 cases).The mRNA and protein levels of CREB were further as- sessed using Western blot and RT-PCR in brain tissues from the patients with brain injury (10 cases) and those with dif- ferent grade gliomas (30 cases). Results The positive rates of CREB immunohistochemistry were 2/5 in control, 10/15 in Ⅰ,Ⅱ, 11/12 in Ⅲ, 28/28 in Ⅳ. The positive rates of CREB were significantly different among different groups (H= 28.183, P〈0.05).The mRNA levels of CREB were 1.00±0.000 in control, 1.35+0.068 in Ⅰ-Ⅱ, 2.88±0.111 in Ⅲ and 3.75〉0.196 in IV. The expression of CREB was higher in the glioma than in control group, and the mRNA levels of CREB were significantly different among different groups (F=l.208,P〈0.05). The protein levels of CREB were 0.311 ± 0.014 in control, 0.469±0.026 in Ⅰ-Ⅱ,, 0.641±0.028 inⅢand 0.896±0.024 in IV. The protein levels of CREB were sig- nificantly different among different groups (F=l.123,P〈0.05). Conclusion The expression of CREB is elevated in glio- mas with different differentiation degrees. The expression of CREB was positively correlated with the degree of differentia- tion, indicating that CREB may have an important regulatory role in the progress of gliomas.
