摘 要:研究了木瓜内生真菌乙酸乙酯萃取部位胃粘膜保护作用及可能的机制.将昆明小鼠完全随机分为空白组、模型组、木瓜内生真菌乙酸乙酯萃取部位(50、100mg/kg)及奥美拉唑(20mg/kg)组,给药组小鼠分别灌胃给予相应药物,每天1次,连续5d,末次给药1h后,除空白组外,各组小鼠分别安10ml/kg灌胃给予阿司匹林的乙醇溶液(15mg阿司匹林溶解于10ml 80%乙醇中),4h后取胃,延胃大弯剖开,除去胃内容物,观察胃粘膜外观形态,并计算胃溃疡发生率、胃溃疡指数及胃溃疡抑制率;刮取胃粘膜,匀浆后,取上清,用试剂盒检测胃粘膜中H+/K+-ATPase、COX-2和PGE2活性;取胃粘膜组织,经HE染色后进行组织形态学观察.木瓜内生真菌乙酸乙酯萃取部位能明显降低胃粘膜中H+/K+-ATPase、COX-2活性,升高PGE2活性,改善胃粘膜组织形态,促进溃疡愈合(P<0.05或P<0.01).结果表明木瓜内生真菌乙酸乙酯萃取部位对胃粘膜损伤有较好的保护作用,其作用机制与其抑制胃粘膜COX-2、H+/K+-ATPase活性和增强PGE2活性有关,然而是木瓜内生真菌乙酸乙酯萃取部位中的何种有效部位/成分起作用及确切的作用机制目前正在进一步研究中.To investigate the protective effect of gastric mucosa of ethyl acetate part of endophytic fungi from Chaenomeles speciosa(sweet)Nakaion on gastric ulcer injury in mice,and to explore its possible mechanism,the experimental mice were randomly divided into control,model,ethyl acetate part of endophytic fungi from Chaenomeles speciosa(sweet)Nakaion(50and 100)and omeprazole groups(20mg/kg),15 mice each group.The gastric ulcers injury were generated by administration of the mixture of ethanol and aspirin which was made up of 10 ml 80% ethanol and 15 mg aspirin at the 4hour after drugs on 5th day.The ulcer index,ulcer inhibition rate,histopathological changes,the H+/K+-ATPase,COX-2and PGE2 activities in gastric mucosa were assessed.The ethyl acetate part of endophytic fungi from Chaenomeles speciosa(sweet)Nakaion(50,100mg/kg)might remarkably decrease H+/K+-ATPase,COX-2activities,and increase PGE2 in gastric mucosa,ameliorate ulcer index,ulcer inhibition rate,histomorphology(P0.05 and P0.01).The ethyl acetate part of endophytic fungi fromChaenomeles speciosa(sweet)Nakaion possess a good protective effect on gastric mucosa injury in mice,inhibiting the activities of H+/K+-ATPase,COX-2,and enhancing PGE2 activity might be one of its molecular mechanisms;but its chemical composition and detailed active mechanisms of toxicity are under studying.
