清脑通络方对AS小鼠血清SOD、MDA、LPO含量及脑组织栓子外渗的影响

摘　　要：目的探讨清脑通络方对动脉粥样硬化(AS)模型鼠脑血管微栓子外渗的影响及其作用机制。方法雄性Apo E基因敲除小鼠高胆固醇饲料饲喂制备AS模型,小鼠随机分为模型组、阿托伐他汀组(3 mg·kg^(-1))、清脑通络方组(30g·kg^(-1))和川芎嗪组(12mg·kg^(-1))。C57BL/6小鼠常规饲料饲喂作为对照组。各组小鼠予相应药物干预4周后,行颈内动脉微栓子注入术,术后48h制备脑组织切片观察栓子外渗率,采血检测血脂水平及血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)、脂质过氧化物(LPO)水平。结果与对照组相比,模型组甘油三酯(TG)、胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、MDA和LPO的含量明显升高,高密度脂蛋白胆固醇(HDL-C)水平、SOD活力及栓子外渗率显著下降;与模型组相比,清脑通络能改善血脂水平,降低MDA和LPO含量,提高SOD活力和栓子外渗率。结论清脑通络方可能通过抗AS所致的氧化应激,一定程度上提高了血管外渗栓子的能力。Objective To study the Qing Nao Tong Luo recipe on cerebrovascular microemboli extravasation and the mechanisms on atherosclerosis（ AS） mice. Methods Male Apo E knockout mice were fed with high cholesterol fee dstuff to create AS model,then they were randomly divided into model group,the atorvastatin group（ 3 mg·kg^（-1））,Qing Nao Tong Luo group（ 30 g·kg^（-1）） and ligustrazine group（ 12 mg·kg^（-1））. C57 BL / 6 mice were fed with regular feedstuff to be control group. Every group was given the corresponding intervention for 4 weeks. Then every mouse was given fluorescent microemboli injection through internal carotid artery,48 hours later,brain slices were obtained for microemboli extravasation observation under confocal microscope,while serum for lipid levels,activity of SOD,MDA and LPO level detection. Results Compared with the control group,the triglyceride（ TG）,total cholesterol（ TC）,low-density lipoprotein cholesterol（ LDL-C）,MDA and LPO level of the model group was increased significantly,high-density lipoprotein cholesterol（ HDL-C） level,SOD activity and embolus extravasation rate decreased significantly. Comparedwith model group,Qing Nao Tong Luorecipe can regulate lipid levels,lower MDA and LPO level,and increase SOD activity and embolus extravasation rate. Conclusion Qing Nao Tong Luorecipe might improve the embolus extravasation ability of vascular through anti-oxidative stress caused by AS,to some extent.
