摘  要:目的优化黄伞胞外多糖发酵培养基。方法在单因素实验的基础上,采用Plackett-Burman实验设计筛选影响多糖产量的培养基成分,然后用最陡爬坡路径逼近最大响应区域,最后通过Box-Behnken设计及响应面分析确定主要影响因素的最佳浓度。结果获得影响多糖产量的培养基成分为:麸皮、黄豆饼粉和磷酸二氢钾。通过Box-Behnken设计及响应面分析确定主要影响因素的最佳浓度为:麸皮11g/L、黄豆饼粉7.3g/L、磷酸二氢钾0.8g/L。结论利用响应面分析法优化黄伞胞外多糖发酵培养基是可靠的,具有实际的应用价值。Objective To optimize submerged cultivation medium components for exopolysaccharide production by Pholiota adipose. Methods Plackett-Burman design was used to screen submerged cultivation medium components affecting exopolysaccharides yield,and then the path of steepest ascent was undertaken to approach the optimal region. Finally the optimum contentration of main influencing factors was determined by Box-Behnken design and response surface analysis. Results Three factors playing the important roles in the medium,including bran,soybean powder and KH2PO4 were selected. By Box-Behnken design and response surface analysis the optimum contentration of main influencing factors were analyzed as: bran 11 g / L、soybean powder 7. 3g / L、KH2PO40. 8g / L. Conclusion It is practical and feasible to optimize submerged cultivation medium components for exopolysaccharides production by Pholiota adipose.
