摘  要:目的观察运动训练和辛伐他汀对肥胖大鼠脂质代谢及PPARa基因、蛋白表达影响。方法采用8周高脂饮食构建肥胖大鼠模型,将造模成功后的49只SD大鼠随机分成安静组(HC,n=12)、给药组(HS,n=12)、运动组(HE,n=12)、给药运动组(HSE,n=13),均喂食高脂高胆固醇饲料。另取同龄SD大鼠10只作为正常对照组(C组),喂食普通饲料。各运动组大鼠采取无负重游泳训练方案,每周运动6 d,每次60 min。辛伐他汀按10 mg/(kg·bw)的剂量进行,每周7 d。干预持续8周后检测各组大鼠体质量、TG、TC、FFA、SOD、MDA、GSH,QRT-PCR方法检测各组大鼠PPARa基因,western-blot方法检测PPARa蛋白表达。结果与C组比较,HC组大鼠体质量显著性升高(P<0.01),血TG、FFA、肝重、肝指数、肝TG、TC、ALT、MDA、PPARa mRNA和PPARa蛋白表达均非常显著性升高(P<0.01),而GSH非常显著性降低(P<0.01);与HC组比较,HS组、HE组、HSE组大鼠除GSH非常显著性升高外(P<0.01),其余指标均显著性降低(P<0.05或P<0.01)。结论 8周游泳运动和辛伐他汀能较好地改善肥胖大鼠因高脂饮食所致的血脂代谢紊乱,降低肝脏中氧化应激,减轻对肝细胞形态和功能的损伤,其中联合干预优于单一干预,具有叠加效应,其机制可能与肝脏中的PPARa基因和蛋白表达变化有关。Objective To observe effects of exercise training and simvastatin on lipid metabolism and PPARa gene and protein expression of obese rats. Methods 49 obese rats model was constructed by fed on a high-fat diet for 8 weeks,then were randomly divided into quiet group(HC,n = 12),drug group(HS,n = 12),exercise group(HE,n = 12),drug plus exercise group(HSE,n = 13),all were fed on high fat and cholesterol. Another 10 SD rats of the same age were chosen as normal control group(group C),and fed on chow. Exercise training was the plan of no load swimming training exercise,6days a week,60 min per day. Simvastatin at 10 mg /(kg·bw) dose,7 days a week. Intervention for 8 weeks later,body weight,TG,TC,FFA,SOD,MDA and GSH were determined,RT-PCR method to detect PPARa genes,and western blot method to detect PPARa protein expression. Results Compared with group C,rats weight group HC increased significantly(P〈0. 01),blood TG and FFA,liver weight,liver index,liver TG,TC,ALT,MDA and PPARa mRNA and PPARa protein expression were very significantly higher respectively(P〈0. 01),and GSH was very significantly lower(P〈0. 01);Compared with HC group,in addition to the GSH was very significant higher(P〈0. 01),the rest of the indicators of rats group HS,HE,HSE were significantly lower respectively(P〈0. 05 or P〈0. 01). Conclusions 8-week swimming and simvastatin can better improve lipid metabolic disorders of obese rats caused by a high-fat diet,reduce oxidative stress in the liver,reduce morphology and function damage of liver cell,especially joint intervention is superior to the single intervention,which mechanism may be related to changes of PPARa gene and protein expression in the liver.
