摘 要:目的分析miR-128在结肠癌细胞中的表达情况,研究其对结肠癌细胞生物学行为的影响及作用机制。方法对转染miR-128模拟物后的结肠癌SW48细胞进行培养,采用MTT法行细胞增殖实验检测细胞增殖能力,细胞划痕法检测细胞迁移能力,Transwell细胞侵袭实验检测细胞侵袭能力;并采用RT-RCR检测miR-128 mRNA的表达,Western blot法检测miR-128蛋白的表达。结果转染miR-128模拟物后的结肠癌SW48细胞较转染空白对照模拟物后的,细胞增殖、侵袭及迁移能力均下降(均P<0.05),且转录因子Sp1基因在mRNA及蛋白水平均下降(均P<0.05)。结论 miR-128可抑制结肠癌细胞增殖及侵袭转移,可能通过调控Sp1基因发挥上述作用。Objective To investigate the expression of microRNA-128 (miR-128) in colon cancer cells and its effect on invasion and metastasis of cancer cells. Methods Human colon cancer SW48 cells were transfected with miR-128 mimics. The cell proliferation was determined by MTT assay, cell migration and invasion were detected by scratch and Transwell methods, the target genes of miR-128 were identified by Western Blot and qPCR methods. Results The proliferation, invasion and migration in miR-128-transfected SW48 cells (P〈0.05). The expression of Spl mRNA and protein level were decreased in miR-128 over-expressing SW48 cells (P〈0.05). Conclusion MicroRNA-128 can inhibit the proliferation and invasion of colon cancer, in which SD1 aene mav be involved.
