胃复方含药血浆对胃癌SGC-7901细胞株增殖及PPARγ基因活性的影响

摘　　要：[目的]通过观察胃复方含药血浆对胃癌SGC-7901细胞株增殖及对PPARγ(过氧化物酶增殖物激活受体γ)的影响,对胃复方含药血浆影响胃癌细胞增殖的可能机制进行探讨。[方法]用不同浓度的胃复方含药血浆处理胃癌SGC-7901细胞,CCK-8法观察其对细胞增殖抑制作用;荧光定量检测法检测PPARγ在胃癌SGC-7901细胞中的表达。[结果]胃复方含药血浆能抑制SGC-7901细胞增殖,作用胃癌SGC-7901细胞株24h后,胃复方空白组、低剂量组、中剂量组、高剂量组OD值分别为0.697±0.040、0.654±0.032、0.548±0.048、0.036±0.029;48h后OD值分别为1.233±0.081、1.131±0.040、0.856±0.055、0.413±0.065;72h后OD值分别为1.736±0.026、1.362±0.013、0.748±0.024、0.326±0.020。差异均有统计学意义,P<0.05。胃复方含药血浆在作用胃癌SGC-7901细胞48h后,PPARγ基因随着浓度的增高表达上调,低剂量、中剂量、高剂量胃复方含药血浆的相对表达量分别是空白血浆的(41.1172±1.9615)倍、(4448.5509±4.7522)倍、(14893.2846±1.0381)倍。(P<0.05)。[结论]胃复方含药血浆可能通过上调PPARγ基因表达的途径来抑制胃癌SGC-7901细胞增殖。[Objective]To explore the possible mechanism of plasma containing Weifufang Decoction on SGC-7901 cell proliferation of gastric cancer by observing the effect of it as well as PPAR gamma gene activity.[Methods]Different concentrations of plasma containing Weifufang Decoction were used on SGC-7901 cells of gastric cancer,and then in the method of CCK-8to observe its inhibitive effects on cell proliferation;Fluorescence quantitative assay was used to detect the PPAR gamma expression in gastric cancer SGC-7901 cells.[Results]The plasma containing Weifufang Decoction can inhibit the proliferation of gastric cancer SGC-7901.After reacted for 24 hours,the OD values of gastric cancer SGC-7901 cell lines in plasma containing Weifufang Decoction blank-control group,low-dose group,middle-dose group,and high-dose group（of gastric cancer SGC-7901 cell lines after 24 hours OD values）were 0.697±0.040、0.654±0.032、0.548±0.048、0.036±0.029;And after 48 hours,the OD values were 1.233±0.081、1.131±0.040、0.856±0.055、0.413±0.065respectively;After 72 hours,the OD values were 1.736±0.026、1.362±0.013、0.748±0.024、0.326±0.020 respectively.The differences were statistically significant（P〈0.05）.After 48 h,Concentration dependence increased the expression of PPAR gamma gene in gastric cancer SGC-7901 cells.The relative expression of low-dose,middle-dose and high-dose of plasma containing Weifufang Decoction was（41.1172±1.9615）times,（4448.5509±4.7522）times and（14893.2846±1.0381）times the level of blank plasma respectively.（P〈0.05）[Conclusion]The plasma containing Weifufang Decoction may improve the（raise）PPAR gamma gene expression level to inhibit the proliferation of gastric cancer SGC-7901 cells.
