摘  要:目的:探讨在氧糖剥夺/复氧糖(OGD/R)情况下腺苷预处理对体外培养星形胶质细胞的影响和脂质运载蛋白-2(LCN-2)表达的影响及其意义。方法:原代培养的大鼠星形胶质细胞随机分为腺苷预处理对照组(Ado—treated contr01)、对照组(Contr01)、实验组(AIX)/R)和模型组(OGD/R)。用MTT法检测各组细胞活力,ELISA检测各组OGD/R后细胞培养基中乳酸脱氢酶(LDH)的浓度,免疫荧光及免疫印迹检测各组LCN-2的相对表达。结果:腺苷预处理对照组与对照组的细胞活力、LDH浓度和LCN-2的表达量均无明显差别。与腺苷预处理对照组、对照组相比,模型组细胞损伤程度和LCN-2的表达量均明显升高。与模型组相比,实验组细胞损伤程度和LCN-2的表达量均明显降低。结论:腺苷可以通过减少LCN-2的释放明显改善OGD/R所引起的星形胶质细胞的损伤,从而增强脑组织对缺血再灌注损伤的耐受。Objective: To discuss the effect of adenosine pretreatment on astrocytes and lipocalin 2 (LCN-2) expression and its significance under oxygen-glucose deprivation and reoxygenation (OGD/R). Methods: Primary cultured rat astrocytes were divided randomly into Ado-treated control group, control group, ADO/R group and OGD/R group. The cytoactivity was detected by MTT assay. The concentration of lactate dehydrogenase (LDH) in the cell medium was indicated by ELISA. The expression of LCN-2 was detected by immunofiuorescence and Western blotting. Results: The cytoactivity, the concentration of LDH and the expression of LCN-2 had no obvious difference between Ado-treated control and control group; The OGD/R group's cell damage degree and the LCN-2 expression increased significantly after the OGD/R compared with Ado-treated control and the control group; The ADO/R group's cell damage degree and the LCN-2 expression level decreased obviously compared with the OGD/R group. Conclusion: Adenosine preconditioning can decrease the ceil damage degree of astrocytes and increase the brain's tolerance to ischemia-reperfusion injury.
