肺腺癌组织和血清中lncRNA-ATB表达及临床意义分析

摘　　要：目的：探讨长链非编码lncRNA？ATB在肺腺癌组织、血清中的表达水平及其临床意义。方法收集本院82例肺腺癌患者的术前血清标本及手术切除的癌组织和配对癌旁组织标本，同时选取年龄、性别配对的65例健康体检者的血清标本为对照，采用实时定量RT？PCR（ qRT？PCR）法检测以上标本的lncRNA？ATB水平；分析肺腺癌组织、血清中lncRNA？ATB水平与临床病理参数（性别、年龄、TNM分期、肿瘤大小、分化程度、淋巴结转移及CEA水平）的关系，采用受试者工作特征曲线（ ROC）评价组织、血清lncRNA？ATB水平在诊断肺腺癌的效能。结果肺腺癌组织lncRNA？ATB水平高于癌旁组织，肺腺癌血清lncRNA？ATB水平高于健康对照，差异均有统计学意义（ P＜0？05），且肺腺癌患者血清与组织lncRNA？ATB水平呈正相关（ r＝0？642， P＜0？05）；肺腺癌组织、血清lncRNA？ATB表达与TNM分期、肿瘤大小及淋巴结转移均有关，其中TNM Ⅱ～Ⅲ期、肿瘤大小＞3 cm和有淋巴结转移者的lncRNA？ATB水平高于对应项（ P＜0？05）；组织、血清lncRNA？ATB水平诊断肺腺癌的价值较好，其曲线下面积和灵敏度均高于CEA。结论 lncRNA？ATB在肺腺癌组织、血清中均为高表达，且与TNM分期、肿瘤大小及淋巴结转移有关，可能在肺腺癌发生发展中有一定作用，且在肺腺癌诊断上，血清lncRNA？ATB水平有较高的价值，可考虑在临床应用。Objective To investigate the expression levels and clinical significance of long noncoding RNA lncRNA activated by TGF β（ lncRNA ATB） in tissues and sera of lung adenocarcinoma. Methods In this study, preoperative serum specimen and sur gical resected cancer tissue together with paired adjacent tissue samples were collected from 82 patients with lung adenocarcinoma. The serum samples of 65 healthy subjects were selected as the control group. LncRNA ATB levels were detected by real time quantitative RT PCR （ qRT PCR） method. The relationship between tissue/serum levels of lncRNA ATB and clinical pathological parameters （ gen der, age, TNM stage, tumor size, degree of differentiation, lymph node metastasis and CEA level） were analyzed. The receiver operat ing characteristic curve （ ROC） was employed to evaluate the efficiency of tissue/serum levels of lncRNA ATB in diagnosis of lung ade nocarcinoma from healthy. Results There were higher levels of lncRNA ATB in both tissues and sera of lung adenocarcinoma as com pared with their controls （ P〈0 05） . The serum level of lncRNA ATB was positively correlated with its tissue level （ r=0 642, P〈0 05）. The expression of lncRNA ATB in lung adenocarcinoma tissues and sera were related to TNM stage, tumor size and lymph node metastasis. The value of tissue and plasma levels of lncRNA ATB for the diagnosis of lung adenocarcinoma was better with a higher are a under the curve and sensitivity than CEA. Conclusion There were higher levels of LncRNA ATB in both tissues and sera of lung adenocarcinoma. Levels of LncRNA ATB were related with TNM stage, tumor size and lymph node metastasis, probably playing a role in development of lung adenocarcinoma. Serum lncRNA ATB has a higher value in the early diagnosis of lung cancer, worthing of clini cal application.
