IL-17体外促进小鼠脾淋巴细胞对髓母细胞瘤的生长抑制作用

摘　　要：目的研究在脾细胞抗髓母细胞瘤的免疫过程中,IL-17所起的免疫调节功能。方法将小鼠脾脏取得的脾淋巴细胞在不同浓度的IL-17下与髓母细胞瘤Daoy细胞株共培养,以Daoy细胞株单独培养组作为对照,用二苯基溴化四氮唑蓝法测定细胞毒性效应。结果实验结果发现各组Daoy细胞株和脾细胞共培养组在不同浓度IL-17（20ng/m L、40ng/m L、60ng/m L、80ng/m L）条件下细胞抑制率有统计学意义（P〈0.01）,且IL-17在一定浓度范围内（20~80ng/m L）,IL-17浓度越高,髓母细胞瘤的细胞生长抑制率越大。此外且随着时间的增加,各组肿瘤细胞的生长抑制率也均有增加。结论 IL-17在一定浓度范围内能增加小鼠脾细胞对髓母细胞瘤Daoy细胞株的生长抑制作用,且在一定时间段内,随着时间的增加,各组肿瘤细胞的生长抑制率也均有增加。Objective To research immune regulation functions of IL-17 in the immune processes of spleen lymphocytes against medulloblastoma. Methods Spleen lymphocytes from mouse＇s spleen were cultivated in IL-17 of different concentrations and Daoy cell strain of medulloblastoma.Single cultivation of Daoy cell strain was set as the control group.Cytotoxic effects were detected by diphenyl tetrazolium bromide method. Results Cell growthinhibiting rate of Daoy cell strain and spleen lymphocytes cultured in IL-17 of different concentrations（20ng/m L,40ng/m L,60ng/m L,80ng/m L） had significantly statistical difference（ P〈0.01）.Within certain range of concentration（20-80ng/m L）,cell growth-inhibiting rate of medulloblastoma was higher as IL-17 concentration grew higher.In addition, growth-inhibiting rate of tumor cells in every group was increased as time went by. Conclusion Within certain range of concentration,IL-17 can increase the growth-inhibiting effect of mouse spleen lymphocytes on Daoy cell strain of medulloblastoma.In addition,within certain time,growth-inhibiting rate of tumor cells in every group increases as time goes by.
