响应面法优化蛹虫草菌液体发酵条件

摘　　要：以蛹虫草菌(Cordyceps militaris)为试材,用单因素试验筛选适宜温度后,利用响应面法优化培养基,最后用优化的条件和培养基进行摇床6d和摇床6d+静置培养10d的液体发酵试验,研究不同培养条件下蛹虫草菌液体发酵后菌丝体产率以及静置培养对虫草素积累量的影响。结果表明:蛹虫草菌适宜温度为25℃,优化培养基为蔗糖4.34%、酵母粉3.06%、硫酸亚铁0.027%、磷酸二氢钾0.2%、硫酸铵0.04%、硫酸镁0.13%、维生素B10.08%、硫酸锌0.06%;摇床6d和摇床6d+静置10d后,发酵液中的菌丝体产率分别为2.568g/100mL和3.389g/100mL,后者菌丝体产率比初始培养基增加了1.73倍,而虫草素积累量分别达到568.329μg/mL和862.893μg/mL,后者比初始培养基中增加了1.56倍。It was experimented to screen suitable temperature for Cordyceps militaris mycelium.After that,liquid medium was optimized by response surface method.Finally,using the optimized conditions,the mycelium was staticly fermented10 days after 6days shaking-flask culture,compared with 6days shaking-flask culture,in order to understand production of Cordyceps militaries mycelium under different culture conditions and the effect of static fermentation on cordycepin production.The results showed that optimal conditions by one-factor experiment were dark at 25℃.Optimal liquid medium by response surface was sucrose 4.34%,yeast extract 3.06%,FeSO40.027%,KH2PO4 0.2%,（NH4）2SO40.04%,MgSO40.13%,vitamin B10.08%,ZnSO40.06%.The yield of mycelium by cordyceps militaris in the optimal medium increased 1.73 times compared with that in initial medium and reached 2.568g/100 mL,while the yield of mycelium reached 3.389g/100 mL with two-stage culture.And the results of two-stage culture showed that the yield of cordycepin increased 1.56 times more that in initial medium and reached 862.893μg/mL,compared with 568.329μg/mL of only 6days shaking-flask culture.
