摘  要:目的探讨Pin2/TRF1结合蛋白X1(Pin X1)在原发性肝细胞肝癌中的表达及其临床意义。方法收集我院43例原发性肝细胞肝癌患者的癌和癌旁正常组织以及病例资料,通过实时荧光定量PCR(Quantitative Real-time PCR)检测Pin X1的m RNA表达水平,并分析表达差异与临床病理特征及患者预后的关系,卡方检验分析率的差异,应用Kaplan-Meier法进行生存分析,Cox风险比例回归模型筛选预后影响因素。结果在43对癌和癌旁正常组织标本中,29例(67.4%)肝癌组织中的Pin X1 m RNA呈低表达,而相应的癌旁组织只有14例(32.6%)呈低表达,差异具有统计学意义(P<0.05);Pin X1 m RNA表达水平与患者年龄具有相关性,生存分析和Cox单因素、多因素风险比例回归模型表明癌组织中Pin X1 m RNA低表达是肝癌患者总生存期的独立预后因子(HR=5.057,P=0.005;HR=3.761,P=0.006)。结论 Pin X1可能是原发性肝细胞肝癌中潜在的抑癌基因,其表达下调在肝癌的发生发展过程发挥重要作用,并可能成为判断肝癌患者预后的重要的生物学指标。Objective To explore the m RNA expression of Pin2 / TRF1-interacting protein X1(Pin X1) in hepatocellular carcinoma tissues as well as its prognostic significance. Methods A total of 43 paired hepatocellular carcinoma specimens and their adjacent normal tissues with corresponding patients '.clinical data were collected..The m RNA expression of Pin X1 was detected by real-time quantitative polymerase chain reaction.(Q-PCR),.and its correlations with pathological features was evaluated by chi-square test,.prognosis were further analyzed by Kaplan-Meier method..Cox proportional hazard model was used to estimate relative risks resulting in a poor prognosis. Results In the whole specimens,.Pin X1 m RNA expression was decreased in 29 tumor tissues.(67.4%),.while only 14 adjacent normal tissues were in low expression with a statistically significance(P〈0.05); there was a correlation between Pin X1 m RNA expression and patient 's age; low m RNA expression of Pin X1 in tumor tissues were crucially associated with patients '.overall survival by univariate and multivariate Cox proportional-hazards regression analysis.(HR =5.057,P =0.005;.HR =3.761,P =0.006),.as the same results as in survival analysis..Conclusion Pin X1 is a potential tumor suppressor gene that inhibits the onset and progression of HCC,.with a prospect that it may be an important biomarker in surveillance of prognosis and treatment.
